摘要
目的观察人抵抗素(Resistin)重组体对胆管癌细胞系QBC939增殖、侵袭、转移及基质金属蛋白酶-2(MMP-2)表达的影响。方法分别采用噻唑蓝(MTT)比色法、Matrigel侵袭实验和迁移实验检测不同浓度的抵抗素对QBC939细胞增殖、侵袭和迁移能力的影响;采用western Blot检测抵抗素对QBC939细胞内MMP-2蛋白表达的影响情况。结果当抵抗素质量浓度高于10ng/nt时,对QBC939细胞增殖具有明显的促进作用(P〈0.01),并呈-定的剂量依赖性;经10、50、100ng/ml抵抗素处理24h后,QBC939细胞体外侵袭及迁移能力均较空白对照组明显增强(P〈0.01);Western blot法检测抵抗素作用QBC939细胞24h后MMP-2表达水平显著增加。结论抵抗素对人胆管癌QBC939细胞的侵袭和转移能力有明显的促进作用,其作用机制可能与提高MMP-2表达水平有关。
Objective To observe the effects of recombinant resisfin(RR)on proliferation,invasion and metastasis and the expression of MMP- 2 of human eholangiecareinoma cell line QBC939. Methods MTr and Transwell were used to analyze the effects of different .concentrations of RR on proliferation, invasion and migration of QBC939 ecUs; the expression level MMP- 2 protein was examined by Western Blot in QBC939 cells. Results RR signitieanfly promoted the proliferation of QBC939 cells when the concentration of RR was higher than 10 ng/ml( P 〈 0.01 ), the effect was dose- dependent in some certain;after treated with 10、 50、 100 ng/ml RR for 24 h,the capacity of irvasion and migration of QBC939 cells were significantly stronger in RR group than in the control group( P 〈 0.01) ; a significant up - regulation of the MMP - 2 protein expression was observed in QBC939 cells treated with resistin for 24 h. Conclusion RR significantly increases the invasion and metastasis capabilities of QBC939 cells in vitro,and the possible mechanism may be related with upregulation of MMP- 2 level.
