摘要
以中国明对虾为试验对象,优化了拥挤应激条件下中国明对虾肝胰脏蛋白质双向电泳的条件,并分析了关联的差异蛋白质。试验结果表明,中国明对虾肝胰脏蛋白双向电泳优化条件为裂解液中添加三丁基膦,采用17cm的固相pH值(pH 4~7)梯度干胶条,上样量为120μg,等电聚焦时间为65 000V/h。蛋白图谱分析结果显示,得到的700多个蛋白点中差异蛋白点11个,其中8个差异蛋白点丰度显著上调(P〈0.05),3个差异蛋白点丰度显著下调(P〈0.05)。研究表明,在拥挤应激条件下中国明对虾肝胰脏蛋白表达发生显著变化。
The two-dimensional electrophoresis(2-DE)was optimized and differentially expressed proteins were investigated in hepatopancreas of Chinese shrimp(Fenneropenaeus Chinensis)under crowded condition.The results showed that the optimal two-dimensional electrophoresis were observed under the conditions of addition of TBP(tributyl phosphate),strips of 17 cm,and pH 4—7IPG(Immobilized pH gradient),loading the sample of 120μg and setting the voltage and power of isoelectric focusing as 65 000 Vhr.The relative 2-DE maps showed that approximate 600 spots was detected in each gel,and a total of 11 differentially expressed protein spots were found,including 8significantly up-regulated protein spots(P〈0.05)and 3significantly down-regulated protein spots(P〈0.05).The findings indicated that proteins in the shrimp hepatopancreas were differently expressed under crowding stress.
