摘要
为建立胃癌小鼠模型,检测T淋巴细胞在免疫系统的水平变化,探讨其在胃癌免疫系统中的作用。采用在615近交系小鼠皮下注射MFC胃癌细胞建立胃癌小鼠模型。流式细胞仪检测胃癌小鼠(n=20)及正常小鼠(n=20)脾脏CD4+CD25+Treg及T细胞亚群水平。结果显示,胃癌实验组小鼠脾CD3+T(42.87±4.33)%和CD8+T(36.55±3.15)%细胞较正常组升高(P<0.01);CD4+T(55.02±2.64)%细胞及CD4+T/CD8+T(1.48±0.13)细胞比值较正常组降低(P<0.01);胃癌实验组小鼠脾CD4+CD25+Treg(2.14±0.91)%细胞较正常组升高(P<0.01)。由此可知,胃癌小鼠脾Treg细胞水平升高,CD4+T降低,CD3+T、CD8+T升高,CD4+T/CD8+T比值降低,可能在胃癌细胞免疫逃逸过程中发挥一定作用。
Mice model of gastric cancer was established to measure the changes of T-lymphocytes level,and to investigate the role of T lymphocytes in the immune system.615 inbred mice were used to establish gastric cancer model by injected MFC cells subcutaneously.The T-lymphocyte subsets and CD4^+CD25^+regulatory T cells(Treg cells) in spleen of gastric cancer mouse(n=20) and normal group(n=20) were assessed by flow cytometry analysis.The level of CD3^+T(42.87±4.33)%,CD8^+T(36.55±3.15)%,CD4^+CD25^+Treg(2.14±0.91)% cells in tumor model was higher than that in the control(P〈0.01).The level of CD4^+T(55.02±2.64)% and ratio of CD4^+T/CD8^+T cells(1.48±0.13) was lower than that in the control(P〈0.01).It can be inferred that elevated levels of Treg cells,CD3^+T,CD8^+T,and decreased CD4^+T and CD4^+T/CD8^+T ratio may be related to immune escape in mice model of gastric cancer.
