摘要
目的 观察丙戊酸(VPA)干预颅脑损伤大鼠后神经功能恢复状况及损伤灶周围脑组织形态学变化,探讨VPA对颅脑损伤大鼠神经功能恢复的影响.方法 将健康SD雄性大鼠90只,随机分成正常对照组、单纯损伤组、VPA治疗组,建立大鼠闭合性颅脑损伤模型并进行干预,采用神经系统疾病严重程度评分(NSS)评价各组大鼠不同时间点的神经功能;应用光学显微镜观察损伤灶周围脑组织形态学的改变.结果 颅脑损伤后1d单纯损伤组和VPA治疗组大鼠NSS评分分别为(12.45 ±0.57)、(12.37 ±0.93)分,均高于正常对照组[(1.55±0.37)分],差异有统计学意义(P<0.05);颅脑损伤后1d单纯损伤组与VPA治疗组大鼠NSS评分差异无统计学意义(P>0.05);颅脑损伤3d后VPA治疗组大鼠神经功能有不同程度的恢复,VPA治疗后3、7、14、21、28 d大鼠NSS评分分别为(10.19 ±0.37)、(8.82±0.71)、(6.42±0.95)、(5.47±0.87)、(3.45±0.89)分,均优于单纯损伤组的(11.61 ±0.34)、(10.47 ±0.93)、(10.29 ±0.37)、(9.82±0.71)、(9.17±0.43)分,差异有统计学意义(P<0.05).病理学检查显示单纯损伤组大鼠损伤灶周围脑组织形态受损明显,VPA治疗组受损较轻.结论 VPA能够通过减轻神经细胞损伤,改善创伤性颅脑损伤大鼠神经功能恢复的程度.
Objective To observe recovery of neural function and changes of morphology in brain tissue of rats with experimental traumatic brain injury,To explore the roles of valproic acid (VPA) in effects on neural function recovery of rats with TBI.Methods Ninty healthy male Sprague-Dawley rats were randomly divided into normal control group,the simple trauma group and VPA treated group.Rat model of closed head injury was established and then interfered by VAP.Neural function of the rats within each group at different time points were evaluated by Nervous system disease Severity Score (NSS).Optical microscope was used to observe the morphological changes of the brain tissue surrounding injury.Results The behavioral scores of rats were more in simple trauma group and VPA treated group (12.45 ± 0.57,12.37 ±0.93) in comparison with normal control group (1.55 ±0.37) one day after experimental brain injury and the difference were statistically significant (P 〈 0.05).No significantly statistical difference was observed between simple trauma group and VPA treated group on 1 day after brain injury (P 〉 0.05).The difference were significant between Nervous system disease Severity Score of VPA treated group (10.19 ± 0.37,8.82 ±0.71,6.42 ±0.95,5.47 ±0.87,3.45 ±0.89) and simple trauma group (11.61 ±0.34,10.47 ±0.93,10.29 ±0.37,9.82 ±0.71,9.17 ±0.43) 3,7,14,21 and 28 d after brain injury (P 〈0.05).Histological examination revealed the injury of brain was obvious in simple trauma group,and the rats in VPA treated group showed alleviated severity of brain injury.Conclusion Valproic acid could improve degrees of neural function recovery of rats with traumatic brain injury by alleviating severity of brain injury.
