摘要
目的探讨黄芩汤(黄芩和甘草)对哮喘模型大鼠白细胞介素-17(IL-17)和白细胞介素-23(IL-23)表达的影响及可能作用机制。方法将32只SD大鼠随机分4组,正常对照组、哮喘模型组、布地奈德组、黄芩汤组,采用卵清白蛋白致敏加激发法复制大鼠哮喘模型。分类计数外周血炎症细胞,采用HE染色评价各组大鼠气道炎症程度,酶联免疫法ELISA检测血清及支气管肺泡灌洗液中IL-17的浓度,免疫组化标记法测量肺部IL-23水平。结果与正常组比较,模型组哮喘大鼠的气道炎症明显加重,血清及支气管肺泡灌洗液中IL-17的水平升高,肺部IL-23的表达增强(P<0.01)。与模型组比较,布地奈德组和黄芩汤组哮喘大鼠的气道炎症明显减轻,血清及支气管肺泡灌洗液中IL-17的水平降低,肺部IL-23的表达下降(P<0.01);结论黄芩汤和布地奈德可以对抗气道炎症且无统计学意义的差异(P>0.05),但前者大鼠体质量增加相对较快,且性情较平稳。
AIM To explore the impact of Huangqin Decoction ( Scutellariae Radix, Glycyrrhizae Radix et Rhi-zoma) on interleukin-17 (IL-17) and interleukin-23 (IL-23) in rats with asthma and its possible mechanism.METHODS Thirty-two Sprague-Dawley rats were assigned to four groups at random: normal control group, asth- ma model group, budesonide group, huangqin group. The model of asthma was established by Ovalbumin sensiti- zing and challenging. The number of inflammatory cells in peripheral was classified and counted. The pathological characters of lung tissues were observed using HE staining. The levels of IL-17 in serum and bronchoalveolar lavagefluid (BALF) were measured by enzyme-linked immunosorbent assay (ELISA). The levels of IL-23 protein in pul- monary tissue were detected by immunohistochemistry. RESULTS Compared with the control group, the airway inflammation in asthma group was significantly aggravateds, IL-17 level in serum and BALF and IL-23 level in pul- monary tissue increased ( P 〈 0.01 ). Compared with the asthma group, the airway inflammation in budesonide group and Huangqin group obviously decreased in severity. Also, IL-17 level in serum and BALF and IL-23 level in pulmonary tissue decreased (P 〈 0. 01 ). CONCLUSION The study shows that Huangqin Decoction and budesonide can relieve airway inflammation with no statistical differences (P 〉 0. 05 ), but the former increases rat body weight more quickly and be of mild disposition.
