摘要
目的 研究周期素依赖性蛋白激酶 5 (CDK5 )的mRNA及蛋白在大鼠发育过程中脑内的表达及分布。方法 采用胚胎至老年期Wistar大鼠脑片行原位杂交组织化学和免疫细胞化学染色。 结果 1 原位杂交结果显示 ,脑内CDK5mRNA在大鼠E14~P35 0整个发育过程中均有表达 ,成年后趋于稳定 ;脑内CDK5mRNA主要定位于神经元 ,阳性区域主要分布于大脑皮层、海马、丘脑、下丘脑、小脑及部分神经核团。 2 免疫组织化学结果显示 ,出生后脑内CDK5蛋白表达较强 ,胚胎及老年鼠表达较弱 ;阳性区域集中在室周区、海马、小脑及部分神经核团内 ;老年鼠仅在海马、小脑蒲肯野细胞层内表达。 结论 以上结果进一步证明 ,脑内CDK5的作用贯穿了整个神经发育的各个时期 ;
Objective To exam the expression and the distribution of the CDK5 mRNA and protein in the developing rat brain. Methods We used embryonic to aged stage Wistar rats by immunocytochemistry staining with anti-CDK5 antibody as well as in situ hybridization technique. Results 1.There were CDK5 specific mRNA expression in the brain tissue during the period from E14~P350, the stable levels of mRNA were detected at adulthood. The location of the CDK5 mRNA was confined to neuronal cell (but not glial) bodies and their processes. The main distribution of CDK5 mRNA was detected in the regions of the cerebral cortex, hippocampus, thalamus, hypothalamus, cerebellum and some neural nucleus mass. 2. The heavy dense signal of CDK5 protein was found in the brain after born, and the signal decrease at embryonic and aged stage. The immuno-positive cells of newborn and adult stages were observed in periventricular zone, hippocampus, cerebellum and some neural nucleus mass. But at aged stage, CDK5 protein expression were found in the neurons in hippocampus and Purkinje cell in cerebellum, while others were not or weakly stained. Moreover, there were marked decrease of CDK5 protein expression in hippocampus at aged stage compared to adult stage.Conclusion These results suggest that CDK5 involved in the whole neurodevelopmental stages of rat brain. A downregulation of CDK5 may be related to the degeneration of learning and memory at aged stage. [
