摘要
目的探讨视网膜母细胞瘤细胞系内抑癌基因Rb1表达下调的分子机制。方法本研究利用real-time PCR和Western blotting等方法分别检测miR-26a和Rb1基因的表达。结果本研究结果显示在人视网膜母细胞瘤组织细胞与正常人视网膜血管内皮细胞间比较,miR-26a表达差异有统计学意义(P<0.005)。miR-26a可以通过与Rb1基因的3'UTR作用而导致人视网膜母细胞瘤细胞内Rb1基因表达下降。结论在Y79 Rb细胞内miR-26a下调抑癌基因Rb1的表达。
Objective To explore the mechanism on down-regulation of Rb1 expression in Rb cells .Methods In our study , the expression of miR-26a and Rb1 was detected by real-time PCR and Western blotting, respectively.Results Here, it was shown that miR-26a was differentially expressed in human retinoblastoma cells and tissues , compared with human retinal microvascular endothelial cells.Our results also showed that miR-26a tightly regulated the expression of Rb1 by specifically targeting a conserved sequence motif in its 3′UTR, resulting in a low expression of Rb1 in retinoblastoma cells .Conclusion The expression of tumor supressor gene Rb1 was down-regulated by miR-26a in Y79 Rb cells.
