摘要
目的:探讨氯沙坦钾对糖尿病肾病(diabetic nephropathy,DN)大鼠肾组织p-JAK2、p-STAT3及VEGF表达的影响。方法:健康雄性SD大鼠30只随机分为正常对照组(C组)、DN模型组(DN组)、氯沙坦钾组(L组)。采用单次腹腔内注射链脲佐菌素(streptozotocin,STZ)法建立DN大鼠模型,周期12周。实验12周末检测大鼠血糖、Scr、BUN、24 h尿蛋白定量;光镜及电镜下观察大鼠肾组织病理改变;采用免疫组化方法检测大鼠肾组织p-JAK2、p-STAT3、VEGF表达。结果:实验12周末,模型组大鼠血糖、Scr、BUN、24 h尿蛋白定量均高于对照组(P<0.05),肾组织中p-JAK2、p-STAT3、VEGF表达均高于对照组(P<0.05);氯沙坦钾组大鼠Scr、BUN、24 h尿蛋白定量均低于模型组(P<0.05),肾组织p-JAK2、pSTAT3、VEGF表达均低于模型组(P<0.05)。结论:氯沙坦钾可能部分通过调控p-JAK2、p-STAT3及VEGF的表达而发挥肾脏保护作用。
Objective:To investigate the effect of Losartan Potassium on p - JAK2,p - STAT3 and VEGF of renal tissue in the diabetic nephropathy rats. Methods:30 SD male rats were divided randomly into control group(group C),diabetic nephropathy group(group DN),Losartan Potassium group(group L). The rats were being intraperitoneal injected with STZ(1% ,45 mg/ kg)to establish diabetic nephropathy rats models,test period 12 weeks. Blood glucose,Scr,BUN,24 - hour urinary protein were measured at the 12th. The renal pathological changes were detected by light microscope and electron microscopy. The expression of p - JAK2, p - STAT3 and VEGF were detected by immunohistochemistry staining sections. Results:After establishing mode for 12 weeks,com-pared with group C,group DN had significant elevated blood glucose,24 - hour urinary protein,Scr,BUN(P ﹤ 0. 05),and immuno-histochemistry staining displayed that the expression of p - JAK2,p - STAT3,VEGF remarkably increased(P ﹤ 0. 05). Compared with group DN,group L had significantly lower 24 urinary protein,Scr,BUN(P ﹤ 0. 05),the expression of p - JAK2,p - STAT3, VEGF were lower than those in group DN(P ﹤ 0. 05). Conclusion:Losartan Potassium may in part through regulating the expression of p - JAK2,p - STAT3 and VEGF play a protective role in the kidney of diabetic nephropathy rats.
