不同频率电针镇痛抗抑郁效应观察和对中枢5-HT的影响 被引量:16

Experimental Observation and Central 5-HT Mechanism on Dual Analgesia and Anti-depression Effect of Electro-Acupuncture with Different Frequencies on Rats

摘要 [目的]观察不同频率电针对健康大鼠的镇痛抗抑郁效应及其与中枢5羟色胺(serotonin,5-HT)的关系。[方法]将SD健康大鼠随机分为空白对照组、2Hz电针组、15Hz电针组、50Hz电针组和100Hz电针组。各电针组大鼠选双侧后肢足三里、昆仑穴进行电针干预1次,电针强度1 mA(1-15min)-2mA(16-30min),时间30min,电针频率分别为2Hz、15Hz、50Hz和100Hz。电针后对各组大鼠进行痛阈、旷场实验测试,用高效液相-电化学法检测各组大鼠的海马和中缝核5-HT含量。[结果]与空白对照组相比,各电针组大鼠痛阈均有上升(P<0.05,P<0.01),2Hz电针组和100Hz电针组抗抑郁效果显著(P<0.001,P<0.05);2Hz电针组海马的5-HT含量上升(P<0.05),100Hz电针组中缝核5-HT含量升高(P<0.05)。[结论]2Hz和100Hz电针均能提高健康大鼠痛阈与抗抑郁能力,但其双重效应的中枢5-HT机制可能不同。 [Objective] To investigate analgesia and anti-depression effect of electro-acupuncture(EA) with different frequencies, and its relationship with serotonin content in central nerve system; [Methods] 60 male SD rats were randomly divided into Control group, 2Hz group, 15Hz group, 50Hz group and 100Hz group.EA(1mA 15min-2mA 15min) was apphed to the bilateral Zusanli(ST36) and Kunlun(BL60) acupoints once for 30 minutes in four EA groups with corresponding frequency of 2Hz, 15Hz, 50Hz and 100Hz. Depression-like behavior in open field test and paw withdrawal latency(PWL) in electronic yon frey test were performed after treatment. The serotonin content in hippocampus and raphe nuclei was tested with high performance liquid electrochemistry(HPLC-ECD). [Results] Compared with the Control group, EA with four frequencies significantly increased the PWL(P〈0.05, P〈0.01); 2Hz and 100Hz EA caused a remarkable anti-depression effect(P〈0.001 ,P〈0.05); the serotonin contents of hippocampus in 2Hz group and that of raphe nuclei in 100 Hz group were notably higher(P〈0.05). [Conclusion] Both 2Hz and 100Hz EA could play dual analgesia and anti-depression effects, and 2Hz EA was especially outstanding in anti-depression. Their central 5-HT mechanism of dual analgesia and anti-depression effects may be different in 2Hz and 100Hz EA.
