温阳通脉方预处理对大鼠心肌缺血再灌注损伤HSP、NF-κB的影响 被引量:4

The Influence of The WenYangTongMai Decoction Preconditioning to HSP and NF-KB in The Ischemia-reperfusion Myocardium of Rats

摘要 目的:观察温阳通脉方预处理对大鼠心肌缺血再灌注损伤的影响,探讨其、保护机制。方法:50只SD雄性大鼠随机分为假手术组、心肌缺血再灌注损伤组(IR组)、心肌缺血预适应组(IP组)、卡托普利组(KT组)、瓜蒌薤白半夏汤组(GL组)、温阳通脉方组(WY组)。建立大鼠心肌缺血再灌注损伤(IR)模型及心肌缺血预适应(IP)模型。检测各组大鼠血清CK-MB、LDH及cTnT含量,免疫组化法检测心肌NF-κB蛋白表达,western-blot检测热休克蛋白(HSP)表达。结果:WY组、GL组、IP组、KT组血清CK-MB、LDH、cTnT含量低于IR组(P<0.05),高于假手术组(P<0.05)。WY组、GL组、IP组、KT组NF-KB表达低于IR组(P<0.05)。WY组、GL组、IP组和KT组HSP70蛋白表达高于IR组(P<0.05)。WY组、GL组、IP组和KT组血清CK-MB、LDH、cTnT含量及心肌NF-κB、HSP表达比较无统计学差异(P>0.05)。结论:温阳通脉方预处理具有心肌保护作用,其保护机制与上调内源性保护因子HSP70,抑制其相关NF-κB蛋白表达有关。 Objective:To study the effect of WenYangTongMai Decoction on rats with myocardial ischemia /reperfusion(I/R) injury and its mechanism. Methods:Fifty male SD rats were randomly divided into sham operation group,ischemia reperfusion(IR)group,ischemia preconditioning(IP) group,captopril(KT) group,Gualouxiebai decoction(GL)group and WenYangTongMai decoction(WY) group. The IR and IP models were established by ligation of LAD. Creatine kinase MB Isoenzyme(CK- MB),lactate dehydrogenase(LDH) and cTnT in serum were measured. The expression of the NF- κB was tested by immunohistochemistry method. The expression of HSP70 was tested by Western blot method. Results:The serum level of CK- MB,LDH and cTnT in IP,KT,GL and WY group were lower than IR group,but higher than sham group. The expression level of NF- κB in IP,KT、GL and WY group were lower than IR group(P〈0. 05). The expression level of HSP70 in IP,KT GL and WY group were higher than IR group(P〈0. 05). but there was no difference in the serum level of CK- MB,LDH,cTnT and the expression level of NF- κB,HSP70 between the treatment groups(P〈0. 05).Conclusion:WenYangTongMai decoction preconditioning can protect ischemia reperfusion rats by uploading the expression of HSP70 and download the expression of NF- κB.
