中等强度持续训练对大鼠骨骼肌线粒体抗氧化能力及ATP酶活性的影响 被引量:1

Effects of moderate-intensity continuous training on mitochondrial antioxidant capacity and activity of ATPase in skeletal muscle:experiment with rats

摘要 目的:观察中等强度持续训练后骨骼肌线粒体抗氧化能力和ATP酶活性的变化。方法 SD大鼠24只,8只为正常对照组(A组),16只采用递增负荷训练建立中等强度持续运动跑台训练模型,又均分为运动2周及6周组(B及C组)。B及C组大鼠按Bradford等的方法分别训练2和6周,每周训练6 d。训练结束后大鼠处死,用差速离心法提取腓肠肌线粒体,紫外-可见分光光度计检测锰超氧化物歧化酶(Mn-SOD)活性;脂质过氧化产物丙二醛(MDA)含量采用TBA(硫代巴比妥酸)法测定;Na+K+-ATP酶和Ca2+Mg2+-ATP酶活性采用酶促反应定磷法测定。结果B组的骨骼肌线粒体Mn-SOD水平与A组相比差异无统计学意义,P>0.05,而MDA含量显著高于对照组,P〈0.05。C组的Mn-SOD水平显著高于A组, P〈0.05,而MDA含量显著低于A组,P〈0.05。B组Na+K+-ATP酶和Ca2+Mg2+-ATP酶活性均显著低于A组,均P〈0.05,而C组的Na+K+-ATP酶和Ca2+Mg2+-ATP酶活性均显著高于A组,均P〈0.05。结论中等强度持续训练一定时间后可以增加骨骼肌线粒体Mn-SOD的活性,降低脂质过氧化水平,减少自由基的损害作用,提高ATP酶活性,改善线粒体能量代谢水平。 Objective To investigate the effects of moderate-intensity continuous training on the mitochondrial antioxidant capacity and activity of ATPase in skeletal muscle so as to provede objective basis for scientific exercise training. Methods Twenty-four SD rats were randomly divided into 2 groups:normal control group (Group A, n=8) and experimental group (n=16). Those rats in the experimental group underwent treadmill exercise at moderate intensity according to the Bredford's model. They were further randomly divided into 2 equal groups to be trained 6 days a week for 2 weeks (Group B) or 6 weeks (Group C). At the end of exercise training all rats were killed with their gastrocnemius muscle obtained and mitochondria therein were extracted by differential centrifugation. Ultraviolet-visible light spectrophotometer was used to examine the activities of manganese superoxide dismutase (Mn SOD) and copper zinc superoxide dismutase (CuZn SOD) . Thiobarbituric acid method was used to detect the content of malondialdehyde (MDA), a lipid peroxidation product. Enzyme reaction phosphorus measurement was used to detect the acxtivities of Na+K+-ATPase and Ca2+Mg2+-ATPase. Results There was no significant difference in the content of Mn-SOD between Group A and Group B ,P>0.05, whereas the content of MDA of Group C was significantly higher than that of Group A ,P〈0.05. The Mn-SOD level of Group C was significantly higher than that of Group A,P〈0.05, however, the content of MDA of Group C was signiuficantly lower than that of Groupo A ,P〈0.05. The activities of Na+K+-ATPase and Ca2+Mg2+-ATPase of Group B were both significantly lower than those of Group A,both P〈0.05, however, those of Group C were both significantly higher than that of Group A ,P 〈0.05. Conclusion A certain period of moderate-intensity continuous training increases the activity of Mn-SOD, decreases the level of lipid peroxidation, reduces the injury of free radical, enhances the activity of ATPase,
