摘要
目的探讨神经突触间隙5-羟色胺(5-HT)神经递质回吸收的抑制剂舍曲林对大鼠各个睡眠时相的影响。方法运用大鼠皮层脑电结合肌电的描记技术,预先给大鼠埋置脑电和肌电电极。采用束缚应激的经典方法进行造模,连续14天,复制大鼠慢性心理应激的模型,监测模型复制以后大鼠12 h的脑电图(EEG)以及肌电图(EMG),通过分析注射舍曲林后大鼠各睡眠时相持续时间及所占睡眠总时间(total sleep time,TST)比例的变化,进而探讨舍曲林对慢性束缚应激大鼠睡眠结构的影响。结果与空白组相比较,模型组大鼠在浅睡眠期(light sleep,LS)、慢波睡眠(slow wave sleep,SWS)以及TST的持续时间均显著缩短(P<0.05),快动眼睡眠(rapid eye movement sleep,REMS)期睡眠的持续时间虽然有缩短的趋势,但是无统计学上的差异(P>0.05)。与模型组相比较,阳性对照组大鼠LS期、SWS期和TST均显著延长(P<0.05),REMS期的睡眠持续时间虽有延长的趋势,但无统计学上的意义(P>0.05);盐酸舍曲林给药组大鼠SWS期和TST均显著延长(P<0.05),LS期和REMS期的睡眠持续时间虽有延长的趋势,但无统计学上的意义(P>0.05)。从各睡眠时相所占TST的比例来看,与空白组相比较,模型组大鼠SWS期所占TST的比例显著减少(P<0.05),而LS期和REMS期睡眠所占TST的比例虽有增加的趋势,但是差异无统计学意义(P>0.05);与模型组相比较,阳性对照组大鼠LS期和REM期睡眠所占比例显著降低(P<0.05),而SWS期所占比例明显增加(P<0.05);盐酸舍曲林给药组大鼠LS期睡眠所占TST的比例显著降低(P<0.05),SWS期所占TST的比例则显著升高(P<0.05),而REMS期睡眠所占比例变化不明显(P>0.05)。结论长期的束缚应激可打乱大鼠正常的睡眠结构,表现为各睡眠时相持续时间的减少以及所占TST比例的变化,而大脑神经突触间隙5-HT神经递质重吸收的抑制剂舍曲林能明显对抗束缚应激对大鼠睡眠造成的影响。
Objective To investigate the effect of sertraline( 5-hydroxytryptamine reabsorption inhibitor) on sleep phases of rats suffering to chronic restraint stress. Methods The EEG and EMG electrodes were implanted to the cortical of rats. The chronic restraint stress method was applied for 14 consecutive days to establish rat psychological stress model. EEG and EMG of rats were monitored by polygraph for 12 consecutive hours in order to analyze the impact of sertraline on the duration and percentage proportion in total sleep time( TST) of each sleep phrase. Results Compared with vehicle,the duration of TST,light sleep( LS) and slow wave sleep( SWS) of rats in model group decreased markedly( P 〈0. 05). Though there was a trend to decrease of rapid eye movement sleep( REMS),the ANOVA did not show significance( P 〈0. 05). The percentage proportion of SWS in TST significantly decreased( P 0.05),while LS and REMS changed slightly( P 〈0. 05). Compared with model group,the duration of LS,SWS and TST of rats in positive control group were increased markedly( P 〈0. 05). The proportion of LS and REMS in TST significantly decreased( P 〈0. 05),while SWS increased markedly( P〈 0. 05). The duration of TST and SWS of rats in administrated group increased markedly( P〈 0. 05). Though there was a porlonged trend on LS and REMS,the ANOVA did not show any significance( P 〈0. 05). The percentage proportion of LS in TST in administrated group significantly decreased( P 〈0. 05),while SWS increased markedly( P〈 0. 05). The percentage proportion of REMS changed slightly( P 〈0. 05). Conclusion According to the change of sleep phase,the structure of sleep is disturbed by chronic mild restraint stress. Sertraline could effectively alleviate the effect of chronic restraint stress on rats.
