摘要
为探讨萝卜过氧化物酶RSPRX1与其抗氧化能力的关系,采用RNAi技术干扰心里美萝卜(Raphanus sativus)过氧化物酶基因rsprx1的表达,并测定萝卜中过氧化物酶活性、过氧化氢酶活性、过氧化氢和可溶性蛋白含量等生理生化指标。结果显示:RNAi可抑制萝卜rsprx1的表达,浸染后18d,过氧化物酶比活力降低幅度最大,较空载体组降低13%。浸染后21d,可溶性蛋白含量降低幅度最大,较空载体组降低14%,此时过氧化氢酶比活力和过氧化氢含量增加最明显,分别较空载体组增加82%和8%。由此,萝卜过氧化物酶基因rsprx1影响其他一些抗氧化物质的合成与代谢,从而影响萝卜的抗氧化能力。
The peroxidase activity,catalase activity,hydrogen peroxide and soluble protein content in R.sativus were determined by studying expression of peroxidase gene rsprx1 by mean of RNAi technique to discuss the relationship between rsprx1 and its antioxidant ability.RNAi can inhibit rsprx1 expression, and the specific activity of peroxidase decreases by 13% compared with non-carrier CK after 18 days’ treatment.The soluble protein content decreases by 14% but the peroxidase specific activity and hydrogen peroxide content increase by 82% and 8% compared with non-carrier CK after 21 days’treatment respectively.In conclusion,rsprx1 influences the antioxidant ability by affecting synthesis and metabolism of antioxidants in R.sativus.
