摘要
目的:探讨醒脑开窍针刺法对脑缺血再灌注大鼠海马细胞骨架调节作用的分子机制。方法:雄性SD大鼠50只,随机分为正常组、假手术组、模型组、针刺组、对照组。采用改进的Longa线栓法制备大脑中动脉梗塞模型。针刺组采用醒脑开窍针刺法,对照组采用传统阳明经针刺法。参考Bederson 6级5分法进行神经功能评分,运用Real-time PCR和Western Blot检测脑缺血后24h大鼠海马MAP-2、NF-L的mRNA及蛋白表达量变化。结果:与模型组相比,针刺组和对照组均可改善缺血再灌注大鼠神经功能缺损症状(P<0.01),针刺组和对照组治疗前后神经功能缺损症状差异具有显著性意义(P<0.01),但治疗后两组神经功能评分比较差异无统计学意义;针刺组和对照组可上调MAP-2、NF-L的mRNA和蛋白表达,与模型组比较差异具有统计学意义(P<0.01,P<0.05),且针刺组优于对照组(P<0.05)。结论:醒脑开窍针刺法与传统针刺法均可上调脑缺血再灌注大鼠海马MAP-2、NF-L的表达,且醒脑开窍针刺法的作用更为明显。
Objective: To investigate the effect of Xingnao Kaiqiao acupuncture method on the molecular mechanism of brain cytoskeleton regulation in cerebral ischemia reperfusion rats. Methods: 50 male SD rats were randomly divided into normal group, sham operation group, model group, acupuncture group and control group. The improved Longa intraluminal thread approach was used to build a middle cerebral artery occlusion model. The acupuncture group was given Xingnao Kaiqiao acupuncture, and the control group was given the traditional meridian acupuncture. The neurological function was assessed with Bederson 6 levels and 5 scores. The mRNA and protein expression changes of MAP-2 and NF-L in hippocampus were detected 24h after cerebral ischemic by using real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) and Western Blot technique. Results: Compared with the model group, the acupuncture group and the control group could improve ischemia reperfusion neurological symptoms (P〈0.01), the difference in neurological symptoms between acupuncture group and control group was significant before and after treatment (P〈0.01), and the difference between the acupuncture group and control group after treatment was not statistically significant, the mRNA and protein expression of MAP-2, NF-L were increased in the acupuncture group and the control group. Compared with the model group, the difference was statistically significant (P〈0.01, P〈0.05), and the difference between the acupuncture group and control group was statistically significance (P〈0.05). Conclusion: The methods of Xingnao Kaiqiao acupuncture and traditional acupuncture may increase the expression of MAP-2, NF-L in cerebral ischemia reperfusion rat hippocampus and Xingnao Kaiqiao acupuncture method is more effective.
