秦巴山区柿叶中单宁的微波提取工艺研究

Optimization of Microwave Extraction Technology for Tannin from the Persimmon Leaf of Qinba Mountain

摘要 利用微波辅助分别以丙酮和含酸无水甲醇为溶剂对柿叶中的单宁进行提取,采用正交试验对提取工艺进行优化。考察了微波功率、反应温度、溶剂浓度、提取时间等因素对单宁提取量的影响。结果表明,以丙酮为提取溶剂时,最佳提取工艺条件为微波功率600 W、反应温度为70℃、溶剂浓度30%、提取时间9 min,由此工艺得单宁的提取量为4.359%;以含酸无水甲醇为提取溶剂时,最佳提取工艺条件为微波功率500 W、反应温度为60℃、溶剂浓度1.4%、提取时间11 min,由此工艺得单宁的提取量为2.957%。以有机溶剂丙酮为提取剂,采用微波辅助提取,单宁提取效果更佳。 Tannin was extracted from the persimmon leaf with microwave extraction and combined with acetone and the methanol as solvent extraction, and the orthogonal test was employed to optimize the process. The effects of concentration of microwave power, microwave temperature, solvent content and extracting time were discussed. The results showed that using acetone as solvent extraction, optimum extraction condition was as follows: microwave power 600 W, microwave temperature 70 ~C, solvent content 30% and extract time 9 rnin, the extraction yield of tannin was 4.359%. Using the anhydrous methanol as solvent extraction, optimum extraction condition was as follows: microwave power 500 W, microwave temperature 60℃, solvent content 1.4% and extract time 11 min. the extraction yield of tannin was 2.957%. Using the acetone as solvent extraction, and combined with microwave irradiation, the extraction yield of tannin was more perfect.
