摘要
目的为探讨脾是否参与胰岛β细胞功能的调节。方法全脾切除小鼠分为3个实验组,另取正常及假手术小鼠作为正常组和假手术组,在实验的3、5、7周,分别取实验1、2、3组及正常和假手术组小鼠采血并取胰腺组织分析相关指标,实验第7周进行糖耐量试验。结果脾切除不影响小鼠空腹血糖和血清胰岛素水平,但实验末期小鼠糖耐量异常,脾切除组葡萄糖曲线下面积(39.38±4.20)与正常组(27.14±5.23)和假手术组(26.36±6.18)相比差异存在统计学意义(P<0.05),胰岛素曲线中,30 min时间点胰岛素水平(31.2±5.66)μIU/mL,显著低于正常组和假手术组[分别为(44.59±4.21)和(46.01±4.38)μIU/mL],差异有统计学意义(P<0.05);免疫组织化学染色显示,随脾切除时间延长β细胞染色面积减少;细胞凋亡的ELISA分析显示,脾切除后5、7周胰岛细胞凋亡聚集值显著升高,与正常组、假手术组比较差异有统计学意义(P<0.05);并且胰岛组织NO水平、iNOS活性及其表达水平与正常组和假手术组小鼠相比均明显升高,差异有统计学意义(P<0.05)。结论全脾切除7周,空腹血糖和血清胰岛素水平不受影响,但糖耐量异常,胰岛β细胞功能减退,与胰岛中iNOS活性及表达增加及NO水平升高引起的β细胞凋亡相关。
[Objective] To explore whether spleen can regulate the function of islet β cell. [Methods] Total splenectomy mice were divided into three groups, normal and sham mice were taken as the normal group and the sham group. At 3, 5, 7 week, the blood and pancreas tissue from group 1, 2 and 3 mice, normal and sham group were collected respectively for analyzing relative data. Glucose tolerance test were also do at 7 week in group 3 and normal and sham group. [Results] Total spleneetomy did not change the levels of free blood glucose and serum insulin, however, caused glucose tolerance abnormal. The area under the glu- cose curve of splenectomy group (39.38± 4.20) were statistically differences from that of normal group (27.14±5.23) and sham group (26.36±6.18) (P 〈0.05). In the curve of insulin, the insulin level of splenectomy mice at 30 min (31.2 ± 5.66 μLU/mL) was lower than that of normal (44.59 ± 4.21) and sham group (46.01 ± 4.38), there were statistically differences (P〈0.05). Immunohistochemical staining showed that staining areas of pancreas islet β cells was gradationally decreased following splenectomy. ELISA analysis of cell apoptosis sug- gested that the enrichment factor of islet cell apoptosis in splenectomy mice rose significantly, compared with that of normal and sham group at 5 and 7 week (P 〈0.05). Meantime, NO levels, iNOS activity and its expression of islet tissue of splenectomy mice significantly increased compared with that of normal and sham group (P〈0.05). [Conclusion] Following total splenectomy for 7 weeks, the concentrations of free blood glu- cose and serum insulin were not influenced, however, glucose tolerance were abnormal, and the function of islet β cell was reduced, which may be associated with islet β cell apoptosis caused by the increasing of iN- OS activity and its expression and of NO levels in the tissue.
