摘要
目的探讨防己黄芪汤对兔肺缺血再灌注后肺水肿的保护作用。方法建立在体兔肺缺血再灌注动物模型。防己黄芪汤组术前连续7 d灌服防己黄芪汤,行左肺缺血再灌注处理;对照组仅行左肺缺血再灌注处理;假手术组只开胸并套带不行缺血再灌注处理。切取各组左肺下叶,部分行组织学观察,部分通过测量肺湿干重的方法计算肺含水量,余肺组织检测超氧化物歧化酶(SOD)含量。最后比较3组肺血管通透性。结果缺血再灌注处理后,对照组肺含水量及伊文思兰含量分别为(89.52±0.36)%和(182.6±17.7)μg/g,明显高于假手术组的(75.57±0.22)%和(113.8±11.5)μg/g;对照组SOD含量(94.32±9.96)NU/mL,明显低于假手术组的(113.52±11.23)NU/mL(P均<0.05),肺组织损伤严重并可见严重的肺水肿。防己黄芪汤组上述指标水平分别为(81.89±0.27)%、(129.3±13.1)μg/g和(126.34±12.56)NU/mL,肺含水量及伊文思兰含量较对照组明显下降,SOD含量明显升高(P均<0.05),组织损伤轻微,肺水肿程度很轻。结论防己黄芪汤通过清除氧自由基、降低肺血管通透性等作用显著减轻了肺缺血再灌注后肺水肿的程度。
Objective To investigate the protective efficacy of Fangji Huangqi Decoction on lung edema after ischemia- reperfusion in rabbits. Methods Establish rabbit lung IRI model in vivo. In Fangfi Huangqi Decoction group, all rabbits were given Fangji Huangqi Decoction for 7 days. In Fangji Huangqi Decoction group and control group, the left pulmonary hilum was occluded and ischemia-reperfusion was done. In sham group, left lung received no ischemia-reperfusion. Left lung tissues in each group were harvested for histopathology analysing. The lung water content was determined and Superoxide Dismutase (SOD) level was checked at the same time. At last, lung vascular permeability were measured. Results After ischemia-reperfusion, the lung tissue water content and Evens blue level in control group were 89.52 ± 0. 36% and ( 182.6 ± 17.7) μg/g respectively, which were significantly higher than those of sham group (75.57 ± 0.22) % and ( 113.8 ± 11.5) μg/g. The SOD level in control group was (94.32 ± 9.96) NU/mL, which was obviously lower than that of sham group ( 113.52 ± 11.23) NU/mL( P 〈 0.05). Those items in Fangji Huangqi Decoction group were ( 81.89 ± 0.27) %, ( 129.3 ± 13.1 )μg/g and ( 126.34 ±12.56) NU/mL; The lung tissue water content and Evens blue level were obviously lower than those of control group; The SOD level was obviously higher than that of sham group ( P 〈 0.05). There is minor tissue damage and degree of pulmonary edema is very light. Conclusion Fangji Huangqi Decoction can significantly prevent lung from lung edema after ischemia-reperfusion in rabbits. The mechanism maybe ascribed to eliminating oxygen free radical and depressing vascular permeability.
