摘要
目的分析miR-27a、miR-30a、miR-664b、miR-451a在肝细胞性肝癌(HCC)患者血清中的表达差异及其与临床病理参数的关系。方法选取HCC患者血清标本66例,相匹配的健康对照血清40例,采用实时定量PCR方法对miR-27a、miR-30a、miR-664b、miR-451a作定量分析。结果相对于健康对照血清,miR-27a、miR-30a、miR-664b在HCC患者血清中的表达差异无统计学意义(P>0.05);miR-451a在HCC患者血清中表达下调,差异有统计学意义(P<0.01),且有淋巴结转移的患者表达水平明显低于无淋巴结转移患者(P<0.05);TNM分期Ⅲ+Ⅳ期的患者表达水平明显低于Ⅰ+Ⅱ期的患者(P<0.05);而miR-451a与患者性别、年龄、肿瘤直径和组织分化程度无关。结论 miR-27a、miR-30a、miR-664b不能为HCC的诊断提供价值;miR-451a在HCC血清中表达显著降低,且与淋巴结转移和临床分期有关,miR-451a可能对HCC的诊断和预后具有潜在的临床应用价值。
Objective To analyze the miR-27a,miR-30a,miR-664b,miR-451a expression differences in hepatocel- lular carcinoma serum and its relationship with clinicopathological parameters. Methods The real-time quantitative polymerase chain reaction (PCR)was used to miR-27a, miR-30a, miR-664b, miR-451a for quantitative analysis in 66 cases of hepatocellular carcinoma and 40 cases of healthy controls, and the relationship of their expressions and clinicopathological parameters were analyzed. Results The miR-27a, miR-30a, miR-664b in hepatocellular carci- noma serum were not seen significant difference compared to healthy control (P 〉 0. 05 ) ; the miR-451 a in hepato- cellular carcinoma serum was down-regulated compared to healthy controls, the difference was statistically significant (P 〈 0.01 ). Furthermore, the expression level of lymph node metastasis was significantly lower than the expression level without lymph node metastasis ( P 〈 0. 05 ) ; the expression level of TNM stage Ⅲ + Ⅳ expression levels was significantly lower than the I + Ⅱ stage expression level ( P 〈 0. 05 ) ; and the miR-451 a was not associated with gender, age, tumor size and histological differentiation degree. Conclusion The miR-27a,miR-30a,miR-664b are considered of no value for the diagnosis of hepatocellular carcinoma. The expression of miR-451 a in serum of hepa- tocellular carcinoma is lower, and is associated with the metastasis and clinical stages and lymph node. MiR-451 a may have potential clinical application value in diagnosis and prognosis of hepatocellular carcinoma
