老年大鼠肝脏内质网GRP78水平揭示其在急性酒精性肝损伤中低应答

摘要 目的 对比观察老年大鼠和青年大鼠肝脏在急性酒精性肝损伤时的GRP78水平变化及其与细胞损伤的关系.方法 通过Real-time PCR技术动态对比测定给予酒精前和后3h、6h、24 h,老年大鼠和青年大鼠肝脏GRP78mRNA表达水平变化,并通过Western-blot法对其蛋白水平进行验证.通过动态测定天门冬氨酸转氨酶(aspartate aminotransgerase,AST)水平,评估酒精对老年大鼠肝脏的损害作用.结果 相较于青年大鼠,老年大鼠发生急性酒精性肝损伤时AST水平更高[(569.5±76.6)U/L vs(218.6±20.5)U/L,P<0.05],并且real-time PCR检测显示老年大鼠的GRP78上调表达时间更慢,幅度更低[(1.69±0.54) vs (295.34±45.72),P<0.01].Westernblot结果印证了老年大鼠的GRP78蛋白水平低于青年大鼠.结论 老年大鼠肝脏GRP78对急性酒精性肝损伤应答更低,细胞损伤更严重. Objective The dynamic levels of GRP78 of endoplasmic reticulum in respond to acute liver injury of alcohol were observed in the ageing rat livers compared to the young controls, and the relationship between the levels of GRP78 and degree of cell injury was checked. Methods The time-sequential comparison of expression of GRP78 between the ageing and the young rats were measured using real time reverse transcript PCR method at the points of 0, 3, 6 and 24 hours after administration of alcohol. The protein levels of GRP78 were detected by Western blotting. The degree ofcell injury induced by alcohol was indicated by the measurement of aspartate transaminase (AST). Results The value of AST was higher in the ageing than that in the young when acute liver injury of alcohol occured. [( 569.5± 76.6 ) U/L vs ( 218.6 ±20.5 ) U/L, P〈0.05] and the RNA level of GRP78 was much lower and later in the ageing than that in the young [(1.69±0.54) vs (295.34±45.72), P〈0.01] by real-time PCR. It was confirmed that the increased range of GRP78 in protein level was lower in the ageing than in the young by Western blot. Conclusion The GRP78 in the aging rat livers responded lower to the acute liver injury of alcohol, while the cell injury was severer than that in the young.
