摘要
目的:观察独活寄生汤对胶原性关节炎大鼠(CIA)膝关节软骨下骨微结构的影响。方法:建立牛Ⅱ型胶原诱导的大鼠关节炎模型,分为模型组、独活寄生汤组和双氯芬酸钠组。采用独活寄生汤灌胃与双氯芬酸钠对照,模型组和正常组采用等量的生理盐水灌胃,持续干预30 d后取材。组织切片苏木素-伊红(HE)染色,测量股骨远端软骨下骨空骨陷窝率,Micro CT扫描观察股骨远端软骨下骨骨密度与骨小梁变化。结果:独活寄生汤组空骨陷窝率减少,与模型组、双氯芬酸钠组比较,差异有统计学意义(P<0.05);独活寄生汤组骨小梁体积比、骨小梁厚度、骨小梁数目均较模型组增加,骨小梁分离度较模型组减少,差异有统计学意义(P<0.05)。结论:独活寄生汤可缓解胶原性关节炎大鼠软骨下骨骨量丢失与骨结构破坏。
Objective: To observe the effect of"Duhuo Jisheng Decoction"on microstructures of subchondral bone in rats with collagen-induced arthritis( CIA). Methods: Type Ⅱ collagen was used to induce the rats,so the models were established and divided into three groups: control group,"Duhuo Jisheng Decoction"group and diclofenac sodium group. We compared the"Duhuo Jisheng Decoction"and diclofenac sodium by intragastric administration. The model group and normal group were treated with the same amount of normal saline for 30 days continuously. Hematoxylin-eosin staining of tissue sections,the empty lacunae rate of subchondral bone in the distal femur was measured. Changes of bone mineral density and bone trabecula in subchondral bone were measured by Micro CT scans. Results: The empty lacunae rate of subchondral bone in the"Duhuo Jisheng Decoction"group was reduced,with a significant difference between the"Duhuo Jisheng Decoction"group and other two groups: model group and diclofenac sodium group( P〈0. 05). Compared with the model group,the bone volume ratio,trabecular thichness,trabecular number in the "Duhuo Jisheng Decoction"group were increased,while the trabecular separation was decreased significantly( P〈0. 05). Conclusion: "Duhuo Jisheng Decoction " can alleviate subchondral bone loss and bone structural damage in rats with arthritis.
