雷公藤内酯醇抑制大鼠脑出血后小胶质细胞和星形胶质细胞的激活 被引量:1

摘要 目的:研究雷公藤内酯醇对大鼠脑出血后小胶质细胞、星形胶质细胞的影响.方法:雄性SD大鼠随机分为干预组、对照组和假手术组.采用Ⅳ型胶原酶肝素生理盐水尾状核立体定向注射法建立大鼠脑出血模型,干预组腹膜腔内注射雷公藤内酯醇,连续7d.在术后每日进行1次Longa神经功能缺损评分.采用免疫组织化学检测小胶质细胞和星形胶质细胞.结果:干预组在术后第2、5、7天的Longa神经功能缺损评分低于对照组;干预组术后第2、7天血肿周边活化的小胶质细胞较对照组明显减少;干预组术后第2天星形胶质细胞较对照组明显减少.结论:雷公藤内酯醇可以部分改善大鼠脑出血急性期神经功能,这可能与其抑制小胶质细胞和星形胶质细胞活化,减轻炎症反应有关. Objective:To explore the effect of triptolide on microglia and astrocyte in intracerebral hemorrhage (ICH) rats.Methods:Male SD rats were randomly divided into an intervention group,a control group and a sham operated group.ICH model was established by microinjecting type Ⅳ collagenase and heparin into rat right caudate nucleus.The intervention group rats were intraperitoneally injected with triptolide everyday for 7 days.Longa neurological defect scores were used for neurological function study on every day after operation.Immunohistochemistry was adopted to assess microglia and astrocyte in the rat brains.Results:Longa neurological defect scores of intervention group were decreased compared with the control group at 2 days,5 days and 7 days after operation; the numbers of microglia and astrocyte were decreased significantly around hematoma in the brains of the intervention group compared with the control group.Conclusion:Triptolide could inhibit activation of microglia and astrocyte and improve partly neurological function in acute ICH rats.
