摘要
目的:本文探讨泽泻汤对梅尼埃病豚鼠模型膜迷路积水的治疗作用及其机制研究,为临床使用本方提供实验依据,并揭示其可能的作用机制。方法:采用腹腔注射醋酸去氨加压素法复制梅尼埃病膜迷路积水模型,以颞骨切片蜗管面积与蜗管面积和前庭阶面积之和的比值、耳蜗的免疫组化AQP-2的表达强度等为指标,研究泽泻汤对其影响。结果:泽泻汤组与模型组的蜗管面积与总面积的比值比较,有显著性差异(P<0.01);免疫组化中AQP-2在泽泻汤组耳蜗表达强度较模型组明显降低。结论:《金匮要略》中的泽泻汤具有显著的减轻膜迷路积水程度的作用,证实了本方治疗梅尼埃病的有效性;并揭示了本方治疗梅尼埃病的作用机制可能是通过直接抑制内耳血管纹、螺旋韧带的AQP-2表达,借助抑制内耳中的AVP-V2R-cAMP途径,从而有利于减轻耳蜗积水。
Objective:To provide experimental reference and reveal the impossible mechanism, based on research on therapeutic effects and mechanism of the Zexie decoction on endolymphatic hydrops in rat models with MD. Methods: Models were made by intraperitoneal injection of desmopressin acetate and the efficacy of the Zexie decoction was researched based on the level of the ratio of temporal bone slices cochlear duct area and cochlear and vestibular canal area, the cochlea immunohistochemical expression of AQP-2. Results: The ratio of the total area of the scroll area in the Zexie decoction group was significantly different from the models group (P〈0.01); the cochlea immunohistochemical expression of AQP-2 in the Zexie decoction group was significantly lower than that in the models group. Conclusion: The Zexie decoction could reduce endolymphatic hydrops and is effective for MD, at the same time, in this paper, it also reveals that the Zexie decoction reduce the cochlea water through direct inhibition of the inner ear vascular pattern, spiral ligament AQP-2 expression, and the inner ear AVP-V2R-cAMP pathway.
