摘要
目的建立厦门市副溶血性弧菌分离株的脉冲场凝胶电泳(PFGE)数据库,分析不同来源菌株的同源性关系。方法 203株菌株的基因组DNA经SfiⅠ酶切、脉冲场电泳后,利用Bionumerics软件分析电泳图谱。结果203株菌株可分成149种不同的PFGE型,91.13%(185株)的菌株具有独特的带型,同时也有少数带型包含的菌株数较多。对于日常监测中分离得到的菌株,PFGE分型结果与水产品的种类、采样时间基本无相关性,79个菌株可分成74个PFGE带型;从历年食物中毒应急监测中分离得到的菌株,其PFGE型相对较少,124株中只有76个PFGE型。应用Bionumerics计算D值为99.81%。结论建立了厦门市副溶血性弧菌的PFGE数据库,为本市副溶血性弧菌的监测和疫情的应急处理提供了科学依据。
Objective To establish pulsed field gel electrophoresis (PFGE)database of Vibrio parahaemolyticus and to analyse the homology during the strains from different sources. Methods The DNA samples of 203 V ibrio para-haemolyticus strains were digested with SfiⅠand then were analyzed by PFGE and bionumerics software. Results 203 strains were divided into 149 distinctive PFGE patterns 91.13% (185 strains) of strains owned unique PFGE pattern, and a few of strains owned more strains.79 strains from surveillance were divided into 74 PFGE patterns,which indicat-ed that there was no relationship between the typing results and aquatic product,sampling time.124 strains from food poisoning were divided into 76 PFGE patterns.The D value was 99.81% calculated by bionumerics software. Conclu-sion The PFGE database of Vibrio parahaemolyticus in Xiamen is established,which providing scientific basis for surveillance and emergency response of Vibrio parahaemolyticus in the city.
