玉米杂交种及其亲本子粒基因差异表达与杂种优势关系的研究 被引量:3

Relationship Between Gene Differential Expression and Heterosis in Kernels of Maize Hybrids and Their Parents

摘要 采用6×5 NCⅡ设计,以玉米乳熟期子粒为材料,在反转录水平上应用mRNA差异显示技术分析玉米杂交种及其亲本间基因差异表达模式,并与杂种优势进行相关分析。结果表明,玉米杂交种与亲本间存在明显的基因差异表达,可分为7种形式。在差异表达模式与杂种表现相关分析中,有15个相关系数达到显著或极显著水平。在差异表达模式与中亲优势相关分析中,株高与杂种特异表达型呈显著正相关;百粒重与单亲表达沉默一型呈极显著负相关;穗粒重和单亲表达沉默一型呈显著正相关;出籽率和蛋白质含量与单亲表达一致二型呈显著正相关;穗粒重和百粒重与杂种特异表达型呈极显著正相关。在与超亲优势相关分析中,株高与单亲表达一致一型呈显著正相关;穗位高和赖氨酸含量与单亲表达一致一型分别呈显著正相关和负相关;行粒数与双亲共沉默型呈极显著正相关,赖氨酸含量与双亲共沉默型呈显著负相关。 The hybrid combinations were derived from 6x5 NC II design and the mRNA differential display technique were used with kernels in milk stage to analyze the expression pattern of gene differences in maize hybrids and their parents. The results showed the genetic expression differences existed in maize hybrid and their parents certainly. It could be divided into 7 types. Named single parent express consistent type Ⅰ (type 110), single parent express consistent typeⅡ (type 011), parents express co-silent type(type 101), single parent express silent type Ⅰ (type 100), single parent express silent type Ⅱ (type 001), hybrid specifically expressing type(type 010) and parents and their hybrids same express type(type Ⅲ). In correlation analysis between differential expression and mid-parent heterosis, plant height and type 010 showed a significantly positive correlation. There was significantly negative cor- relation between 100-kernel weight and type 100. There was significantly positive correlation between spike grain weight and type 100. Seed-producing percentage and protein content showed significantly positive correlation with type 011. Spike grain weight and 100-kernel weight showed significantly positive correlation with type 010. In correlation analysis between differential expression and heterobehiosis, there was significantly positive correlation between plant height and type 100. There was significantly positive correlation between ear height and type 110, there was negative correlation between lysine content and type 110.
