摘要
目的研究ZNF217在宫颈癌、宫颈上皮内瘤变及正常宫颈组织中的表达,探讨ZNF217在宫颈癌的发生、发展中的作用,为宫颈癌的临床诊断和治疗提供重要的理论依据。方法采用免疫组织化学SP法检测ZNF217在60例宫颈癌(宫颈癌组)、18例低度宫颈上皮内瘤变(CINⅠ组)、32例高度宫颈上皮内瘤变(CINⅡ~Ⅲ组)和20例正常宫颈组织(正常组)中的表达。结果①ZNF217蛋白主要表达于宫颈上皮细胞浆。正常宫颈组织、CINⅠ、CINⅡ~Ⅲ及宫颈癌中的ZNF217蛋白的阳性表达率分别为25.0%、33.3%、50.0%、81.5%。宫颈癌组与正常宫颈组、CINⅠ组与宫颈癌组、CINⅡ~Ⅲ组与宫颈癌组间ZNF217阳性表达率有显著差异(x^2值分别为18.755、12.874、7.762,均P<0.01),而正常宫颈组与CINⅠ组、与CINⅡ~Ⅲ组ZNF217阳性表达率无明显差异(x^2值分别为0.320、1.299,均P>0.05)。ZNF217在宫颈癌组织中的表达与组织分化程度和淋巴结转移有关(x^2值分别为12.851、6.222,均P<0.05),但与组织类型和临床分期无关(x^2值分别为0.009、6.675,均P>0.05)。结论凇17高表达在宫颈癌的发生发展中起了重要作用。
Objective To detect the expressions of zinc finger protein 217 (ZNF217) in human normal cervical epithelium, cervical intraepithelial neoplasia (CIN) and cervical carcinoma to investigate the roles of ZNF217 in the carcinogenesis and development of cervical carcinoma, and to provide theoretical basis for clinical diagnosis and therapy. Methods The expressions of ZNF217 were detected by immunohistochemistry SP method in 60 patients with cervical carcinoma (cervical carcinoma group), 18 cases with low grade of cervical intraepithelial neoplasia (CIN I group), 32 cases with high grade of cervical intraepithelial neoplasia (CIN Ⅱ~Ⅲ group) and 20 cases with normal cervical epithelium (normal group). Results ZNF217 was mainly located in cervical cytoplasm. The positive-expression rate of ZNF217 was 25.0%, 33.3% , 50.0% and 81.5% in normal cervical epithelium tissues, CIN I tissues, CIN Ⅱ~Ⅲtissues and the tissues of cervical carcinoma, respectively. There was significant difference in the positive-expression rate of ZNF217 between cervical carcinoma group and normal cervical epithelium group, between CIN I group and cervical carcinoma group, between CIN Ⅱ~Ⅲ group and cervical carcinoma group (X2 value was 18. 755, 12. 874 and 7. 762, respectively, all P 〈 0.01 ). But there was no significant difference between normal cervical epithelium group and CIN I group, between normal cervical epithelium group and CIN Ⅱ~Ⅲ group (X2 value was 0.320 and 1. 299, respectively, both P 〉 0. 05). In the cervical carcinoma group, the expression of ZNF217 was correlated with histological grade and lymphatic metastasis (X2 value was 12.851 and 6. 222, respectively, both P 〈 0.05 ), but it was not correlated with histopathological type and clinical stage (X2 value was 0. 009 and 6. 675, respectively, both P 〉 0.05 ). Conclusion High expression of ZNF217 plays an important role in the carcinogenesis and development of cervical carcinoma.
