摘要
为满足栽培和保护资源的需要,以叶柄段为材料,进行了愈伤组织生长和分化培养、生长芽试管苗培养及移栽与移植的研究,成功获得了生长旺盛的试管苗。结果表明:2/3 MS+6-BA0.5 mg·L-1+蔗糖36g·L-1+2,4-D2.0 mg·L-1是叶柄段愈伤组织生长培养的适宜培养基;1/2MS+琼脂0.5%+蔗糖26g·L-1+6-BA0.8 mg·L-1+NAA0.1 mg·L-1+IAA0.1 mg·L-1是愈伤组织分化培养的适宜培养基;1/3MS+琼脂0.5%+蔗糖16g·L-1+IAA 0.4 mg·L-1是生长芽生根培养的适宜培养基。试管苗移栽成活率为92.4%,移植成活率为97.4%。移植的试管苗能使野生鲜黄连的植物学性状完全得到保持。
To meet the needs of cultivation and protection of wild resource, with the petiole segments of Jeffersonia dubia Maxim. as materials to study the induction and differentiation of the callus, growth rooting of buds, transplanting and colonization of test-tube seedling. The results showed: 2/3MS +6- BA0.5 mg·L-1+ sucrose36g ·L-1+2,4--D2.0 mg ·L-1 was suitable medium of petiole segment's callus culture; 1/2MS +agar0.5 % +sucrose26g·L-1+6 -BA0.8 mg·L-1+NAA0.1 mg ·L-1+IAA0.1 mg·L-1 was suitable medium for callus differentiation culture; 1/3MS +0.5 % agar + sucrose16g ·L-1 +IAA 0.4 mg·L-1 was suitable medium for growth rooting of buds ; the survival rate of test-tube seedling' s transplanting was 92.4 % and the colonization survival rate was 97.4 %. The test-tube seedling maintained botanical traits of wild Jeffersonia dubia Maxim.
