美洲大蠊提取物制备工艺初步研究 被引量:1

摘要 目的开发一种标准化的美洲大蠊提取物制备方法。方法通过单因素实验观察不同盐离子浓度、pH和提取时间对美洲大蠊提取物蛋白提取的影响,采用正交设计试验对提取工艺进行优化。结果各因素对美洲大蠊蛋白提取效率影响大小的顺序为:盐浓度>时间>pH。其中,盐浓度对提取物中蛋白浓度的影响最大(P<0.05)。美洲大蠊提取物的最佳提取条件为:提取液为磷酸盐缓冲液,盐浓度为0.4mol/L,pH为8.0,提取时间为6h。结论成功开发出一种美洲大蠊提取物的标准化提取方法,为研究美洲大蠊过敏机制及其诊断与治疗提供了有利工具。 Objective To develop a standardized procedure for preparing American cockroach extract. Methods The influences of various parameters, such as the time of extraction, concentration of sodium chloride(NaCl), and pH value of sample, on the efficiency of protein extraction were evaluated. The extraction procedure was then optimized using orthogonal test. Results The influences of various parameters on the efficiency of protein extraction were in an order of concentration of NaC1 〉time of extraction〉pH value, of which the concentration of NaC1 was the most(P〈0. 05). The conditions for the protein extraction were NaC1 concentration of 0.4 mol/L, extraction time of 6 hours and pH 8.0. Conclusion A standardized procedure for preparing American cockroach extract has been developed successfully, which will be useful for the diagnosis and management of cockroach allergy.
