摘要
目的:以肌注氢化可的松建立肾阳虚大鼠模型为研究平台,通过血管紧张素Ⅱ(AngⅡ)、血管紧张素受体1(AT1R)指标探讨温补肾阳法对肾阳虚模型大鼠体液调节系统的影响,也为肾主水理论寻找科学依据。方法:60只健康雄性SD大鼠,随机分为对照组、模型组、干预组,对照组不造模,给予生理盐水灌胃[10ml/(kg·d)];模型组注射氢化可的松[2ml/(kg·d)],造模后给予右归胶囊混悬液灌胃[10ml/(kg·d)],干预组造模后给予生理盐水灌胃[10ml/(kg·d)]。第14天三组各处死10只,第42天,各组再处死剩余的大鼠,取肺脏组织常规切片,免疫组化法检测AngⅡ、AT1R在肺脏的表达。结果:免疫组化法检测AngⅡ、AT1R蛋白阳性反应产物呈现棕黄色,第14天,与对照组比较,模型组和干预组大鼠肺组织AngⅡ表达减少,AT1R表达也减少,差异有统计学意义(P<0.05);第42天时,与模型组比较,干预组大鼠肺组织AngⅡ和AT1R表达较多,差异有统计学意义(P<0.05)。结论:肾阳虚大鼠肺组织AngⅡ、AT1R的表达明显下降,表明肾阳虚大鼠体内体液代谢血液循环受到影响;温补肾阳名方右归胶囊可以通过增加模型大鼠肺组织AngⅡ和AT1R的表达,从而缓解肾阳虚对模型大鼠体液调节系统紊乱的影响。
Objective: To study the effect of prescription for warming and recuperating the kidney-yang on angiotensin receptor (Ang Ⅱ ) and angiotensin receptor 1 (AT1R) in hydrocortisone induced rat model of kidney yang deficiency. Methods: Sixty healthy male SD rats were randomly divided into control group, model group, and Yougui capsule intervention group. Immunohistochemistry was used to detect the expression of Ang Ⅱ and ATIR in lung tissue. Results: Both Ang Ⅱ and AT1R expression levels in lung were lower in model group and intervention group of than in control group after 14 days (P〈0.05). But after 42 days' treatment, both Ang Ⅱ and ATIR ex- pression levels were higher in Yougui-intervention group than in model group (P 〈 0. 05). Conclusion: The prescription for warming and recuperating the kidney-yang could regulate the dysfunction of kidney yang deficiency rats through up-regulating the expression of Ang Ⅱ and AT1R.
