摘要 目的探讨体外实验中蒲公英提取物改善胰岛素抵抗的量-效与时-效关系。方法采用地塞米松诱导3T 3-L1脂肪细胞建立胰岛素抵抗模型。设立空白对照组、地塞米松模型组、阳性对照组(罗格列酮终浓度为3.4μg/ml),不同剂量的蒲公英提取物(终浓度分别为0.1、0.3、1、3、10、30μg/ml)组,观察各组在有胰岛素参与和无胰岛素参与情况下,在不同时间节点(24、48、72、96 h)培养基中葡萄糖浓度的变化。结果与空白组和模型组比较,在有胰岛素与无胰岛素情况下蒲公英提取物各剂量组培养基中葡萄糖含量均下降,且呈时间与剂量依赖性。结论蒲公英提取物改善胰岛素抵抗的效应呈较好的量-效与时-效关系。 Objective To investigate the effect of dandelion extracts on insulin resistance in vitro.Methods Insulin resistance was induced by dexamethasone in 3T3-L1 adipocytes.The cells were treated with 3.4 μg/ml rosiglitazone(positive control),different concentration of dandelion extracts(0,0.1,0.3,1,3,10 and 30 μg/ml) for 24,48,72 and 96 h.The change of glucose concentration in culture-medium of the different treatment groups with or without insulin at different time points were measured.Results The glucose concentrations in culture-medium of dandelion extracts-treatment groups with or without insulin were decreased in a time-dependent and dose-dependent manner.Conclusion Dandelion extracts could lessen insulin resistance,and it suggests preferable dose-effect and time-effect manner.
