姬松茸多糖对T淋巴细胞表面分子表达及γ干扰素分泌的影响 被引量:5

摘要 目的:研究姬松茸中分离纯化的低分子量多糖(ABP-AW1)促进卵清白蛋白(ovalbumin,OVA)抗原诱导小鼠脾淋巴细胞亚群效应细胞的应答格局的影响。方法:ABP-AW1和OVA皮下两点免疫ICR小鼠,于0,15 d各免疫一次,第28天分离培养脾淋巴细胞,MTT法测定脾淋巴细胞增殖,FCM(flow cytometry)分析免疫鼠脾中T淋巴细胞亚群CD4+T和CD8+T的百分含量;免疫鼠脾淋巴细胞体外给予同种抗原再刺激24 h,分析ABP-AW1对脾CD69+CD4+T淋巴细胞的表达及胞内因子IFN-γ+CD4+T的调节作用。结果:ABP-AW1能增强OVA诱导的Th淋巴细胞亚群CD4+T细胞,CTL淋巴细胞亚群CD8+T细胞增殖;ABP-AW1也能增进CD4+T细胞早期活化标志CD69+百分比增加,诱导CD4+T胞内Th1型细胞因子IFN-γ分泌水平增多。结论:ABP-AW1佐剂上调脾脏中淋巴细胞亚群的增殖,特别是Th1细胞的募集和活化,研究结果为设计理想的免疫佐剂提供了基础研究数据资料。 OBJECTIVE To discover the effect of ABP-AW1 on effector ceils of splenocytes subsets of mice induced by OVA immunized mice,and the low molecular weight polysaccharide (ABP-AWl) isolated from the mushroom of Agaricus blazei Murill. METHODS ICR were subcutaneously immunized twice with OVA mixed in ABP AWl on Day 1 and Day 15. Spleno- cytes were acquired on Day 28, splenocyte proliferation was measured by MTT. Positive percent of CD4+ and CD8+ Tcell in pe- ripheral blood was measured using FACS. Splenocytes derived from immunized mice were restimulated in vitro with OVA, after incubated for 24 h, the expression of splenocytes surface CD4,CD69 (T cell early activation marker) and intracellular cytokine IFN-y were assayed with FCM. RESULTS ABP-AW1 significantly up-regulated the positive percent of CD4+ and CD8+ T Jym- phocytes both in spleem the ratio of activated T cells (CD69+ CD4+ ) to CD4+ T cells in ABP-AWl immunized was higher than OVA alone,The proportion of IFN-y+ CD4+ T double-positive cells from mice administered ABP-AWl was markedly increased also. CONCLUSION ABP-AWl can promote protein antigen induced proliferation of lymphocyte subsets, especially promotes development of Thl polarization. Therefore, using ABP-AWl as adjuvants in vaccine will be promising.
