摘要
目的探讨凋亡肿瘤细胞抗原负载的树突状细胞(DC)对喉癌细胞杀伤作用的影响。方法用细胞因子重组人白细胞介素4(rhIL-4)、重组人粒细胞巨噬细胞集落刺激因子(rhGM-CSF)与肿瘤坏死因子α(rhTNF-α)体外诱导扩增人外周血单个核来源的DC,6 d后将DC分为凋亡组、冻融组和未处理组;各组细胞继续培养4 h后,分别采用流式细胞术检测DC表面分子的表达、ELISA法检测干扰素γ(IFN-γ)的水平、MTT法评估各组DC刺激细胞毒性T淋巴细胞(CTL)增殖的能力及诱导CTL的杀伤活性。结果与其余各组相比,凋亡组DC表面分子与IFN-γ的表达明显增加(P<0.05),CTL的增殖能力与杀伤活性增强(P<0.05)。结论凋亡肿瘤细胞可以促进DC的分化与成熟,并能使DC产生高效而特异性的抗喉癌免疫应答。
Objective To explore the killing effect of apoptotic-tumor-cell-loaded dendritic cells( DCs) on the laryngeal carcinoma cells. Methods DCs from peripheral blood monocytes were induced by recombinant human interleukin-4( rhIL-4),recombinant human granulocyte/macrophage colony-stimulating factor( rhGM-CSF) and tumor necrosis factor( TNF-α) in vitro,then were divided into the apoptotic laryngeal carcinoma cells group,freeze-thaw cells group and untreated cells group 6 days later. After continuous culture of DCs for 4h,the expressions of the surface markers of DCs were detected by flow cytometry,the level of Interferon-γ( IFN-γ) was measured by ELISA,and the proliferation ability and killing effect of cytotoxic T lymphocyte( CTL) stimulated by DCs were evaluated by MTT. Results Compared with other groups,apoptotic laryngeal carcinoma cells group could enhance the expression of the surface markers CD83,CD86,HLA-DR and IFN-γ( P〈 0. 05),and the proliferation ability and killing effect of CTL increased( P〈 0. 05). Conclusion Apoptotic carcinoma cells can promote the differentiation and maturation of DCs,and induce effective and specific immune response against laryngeal carcinoma.
