摘要
目的:探讨温阳化瘀方对糖尿病周围神经病变大鼠NGF和IGF-1及神经传导速度的影响。方法:取SPF级雄性大鼠,随机分为正常对照组,高脂高糖(模型)组,弥可保组,二甲双胍(降糖)组,温阳化瘀方(试药)大、中、小剂量组。除正常对照组外,各组大鼠禁食12 h,按60 mg/kg剂量尾静脉一次性快速注射链脲佐菌素(STZ)制备动物模型,并以高脂高糖饲料,于模型制作当日各给药组大鼠分别ig给予药物治疗,每日1次,连续12周,其中降脂组给予立普妥1.25 mg·kg-1,降糖组给予二甲双胍670 mg·kg-1,试药大剂量组给予温阳化瘀方浓缩液18 g·kg-1,试药中剂量组给予温阳化瘀方浓缩液12 g·kg-1,试药小剂量组给予温阳化瘀方浓缩液6 g·kg-1。正常对照组、模型组给予同样量的生理盐水。每10 d称量各组动物体重1次;给药最后1 d取血测定空腹血糖;取血清采用酶联免疫吸附分析方法测大鼠血清NGF和IGF-1。坐骨神经神经传导速度测定,用刺激电极法。结果:温阳化瘀方对DPN大鼠血清NGF、IGF-1有一定调节作用,与模型组比较,大、中、小3个剂量组均显示有非常显著的差异(all P<0.01)。温阳化瘀方对DPN模型大鼠所致坐骨神经传导速度减慢组有明显的治疗作用,与模型组比较,小剂量组在前6周有显著差异(P<0.05),但从第9周开始有非常显著的差异(P<0.01);而大剂量组从第3周开始,就显示具有非常显著的差异(P<0.01)。结论:温阳化瘀方能促进NGF、IGF-1的增长,改善DPN所致传导速度。显示本方对DPN有一定治疗作用。
Objective :To investigate the effects of Wenyang Huayu Decoction on nerve growth factors (NGF), insulin growth factor (IGF) -1 and motor nerve conduction velocity (MNCV). Method: The male SPF level rats were randomly divided into control group,high fat and sugar (model) group,high fat and sugar + lipitor ( lipid - lowering) group,high fat and sugar + methylcobalamin group, high fat and sugar + treated with metformin (hypoglycemic) group, high fat and sugar treated with Wenyang Huayu Decoction large dose, middle dose and small dose groups. The rats of every group were fed with high fat and high sugar food ,fasting for 12 h. Intravenous injection of streptozotocin 60 mg/kg was used to prepare the animal model except control group. In addition to the normal control group, rats in other groups were fed with high fat and high sugar food. In addition to the normal control group, other groups and model rats were given drug treatment, once a day,for 12 weeks ,weighing each 10 d. Among them,the rats in lipid - lowering group took 1. 25 mg/kg lipitor;hypogly- cemic group rats took 670 mg/kg metformin; Wenyang Huayu Decoction large dose group, middle dose group and small dose group rats took 18 mg/kg, 12 mg/kg and 6 mg/kg Wenyang Huayu Decoction. Control group and model group rats took normal salt comadministration by gavage by equal content. The levels of serum NGF and insulin growth factor (IGF) - 1 were detected by enzyme - linked immunosorbent assay (ELISA). The motor nerve conduction was detected with electrode. Results : Wenyang Huayu Decoction can definitely adjust the effect on serum NGF and IGF - 1 in DPN rats ( all P 〈0.01 ). The decoction could increase MNCV while MNCV was slowed down. Small dosages were formed in notable difference of the first 6 weeks (P 〈0.0.5). But it had very notable difference from 9'h week (P 〈0.01 ).
