摘要
目的:建立简便和灵敏的双试剂自动化分析法测定血清D-3羟丁酸。方法在NAD和D-3羟丁酸脱氢酶存在下,用氯化硝基四氮唑兰(NBT)作为显色剂测定血清中的D-3羟丁酸。结果该法线性范围为0~7.0mmol/L,平均回收率为98.7%,批内变异系数和批间变异系数均小于2%,与对照试剂盒比较有良好的相关性(r2=0.996),具有良好的抗干扰能力。150例健康人血清D-3羟丁酸含量为0.03~0.30mmol/L。结论 NBT自动分析法测定D-3羟丁酸方法简便、灵敏可靠、适合临床应用。
Objective To establish a simple and sensitive automatic method for determination of D-3-hydroxybutyric acid in serum with two reagents.Methods With the existence of NAD and D-3-hydroxybutyrate dehydrogenase, D-3-hydroxybutyric acid in serum was determined by NBT as a color-developing agent. ResultsThe linearity range of the method was 0-7.0 mmol/L, average recovery rate was 98.7%, and both the intra-assay and inter-assay coefficients of variation were less than 2%. There was a good correlation compared with the reference kit (r2=0.996), and the capacity of resisting interference was good. The concentration of D-3-hydroxybutyric acid in the serum of 150 healthy people was 0.03-0.30 mmol/L.Conclusion The NBT automatic analysis method for determination of D-3-hydroxybutyric acid is simple, sensitive and reliable, which is appropriate to be applied clinically.
