摘要
目的:比较体外培养人子宫肌瘤细胞组织块法和胶原酶消化法的优缺点,探索子宫肌瘤细胞原代培养的最佳方法,为在子宫肌瘤生理和病理等方面的研究提供一种更有效、简便的细胞培养技术。方法:采用组织块法和胶原酶消化法对人子宫肌瘤组织进行体外培养,运用光学倒置显微镜、HE染色观察细胞生物学特性,免疫荧光法进行细胞鉴定。结果:倒置显微镜及HE染色观察两种细胞的形态均多成梭形,平滑肌肌动蛋白免疫荧光染色均呈阳性。结论:组织块培养法与胶原酶消化法作为人子宫肌瘤细胞体外培养的两种方法,均可成功建立子宫肌瘤细胞有限细胞系。通过比较发现,消化法虽相较于组织块法方法稍复杂,但具有短期内可获得大量细胞且细胞纯度较高的优势。
Objective: To compare the advantages and disadvantages of tissue piece method and collagenase digestive method during in vitro culture of human uterine fibroids cells, explore the optimal method of primary culture methods of human uterine fibroids cells, provide a more effective and simple cell culture technique for physical and pathological studies about uterine fibroids. Methods: Tissue pieee method and eollagenase digestive method were used for in vitro culture of human uterine fibroids cells respectively, cell biological characteristics were observed after HE staining under inverted optical microscope, cell determination was performed using immumofluorescence method. Results: Human uterine fibroids cells cultured by the two methods were fusiform after HE staining under inverted optical microscope, immunofluorescent staining of actin in uterine smooth muscle was positive. Conclusion : Both tissue piece method and collagenase digestive method can successfully establish uterine fibroids cell line; compared with tissue piece method, digestive method is more complicated, but it has the advantages of obtaining a large number of cells and high purity in a short time.
