摘要
目的构建大鼠氯化锂-匹鲁卡品颞叶癫痫模型,观察海马和颞叶皮质中pleiotrophin表达的动态变化。方法选取清洁级SD大鼠幼鼠(48只)进行随机对照试验,采用随机数字法分为2组:实验组(24只)和对照组(24只),对照组、实验组分别又均分为2 h模型、3周模型、8周模型3组,每组8只。所有幼鼠腹腔注射氯化锂(127 mg/kg),24 h后,模型组腹腔注射匹鲁卡品(30 mg/kg),每30 min给予匹鲁卡品(10 mg/kg)腹腔注射,直到出现癫痫持续状态,对照组幼鼠腹腔注射等剂量生理盐水,90min后观察大鼠的行为变化,并用免疫组化及Real-time PCR方法检测癫痫后不同时间点脑内pleiotrophin表达的变化。结果对照组均未出现癫痫发作,模型组大鼠在给予匹鲁卡品90 min后均达到Ⅴ~Ⅴ级发作,达到癫痫持续状态。匹鲁卡品诱发癫痫后2 h^8周颞叶皮质和海马中pleiotrophin表达显著上升,颞叶皮质和海马中对照组2 h、3周、8周分别与实验组2 h、3周、8周组比较差异有统计学意义(P<0.01)。结论癫痫后大脑颞叶皮质和海马pleiotrophin表达明显增加并且持续很长时间。
Objective To build temporal lobe epilepsy models by chloride-pilocarpine method in rats,so as to observe the dynamic expression of pleiotrophin in the hippocampus and cortex of temporal lobe in rats.Methods Totally 48 SD young clean grade rats were randomly divided into two groups,experimental group and control group,and each group was divided into 3 sub-groups of 2 hours,3 weeks and 8 weeks (each subgroup has eight rats).All rats were intraperioneally injected with lithium chloride at a dose of 127 mg/kg; 24 hours later,the rats in model group were injected with pilocarpine (30 mg/kg,intraperioneally) and the dosage was added 10 mg/kg every 30 minutes till status epilepticus occurred,and the control group rats were received normal saline; behavioral changes were observed 90 minutes later.Immunohistochemisay and Real-time PCR analysis were performed to detect the dynamic expression of pleiotrophin in the rat brain at different time points after epilepsy.Results Significantly increased expression of pleiotrophin was observed in the hippocampus and cortex from 2 hours to 8 weeks after epilepsy.There are significant differences in the temporal lobe and hippocampus between the control groups of 2 hours,3 weeks,8 weeks and the experimental groups,respectively.Conclusion The significantly increased expression of pleiotrophin in the hippocampus and cortex of temporal lobe after epilepsy of rats last for a long time,which provides a new thought for epilepsy treatment by revealing the mechanism of increased pleiotrophin expression following epilepsy.
