地肤子提取物毒杀螨的蛋白组双向电泳分析

摘要 山楂叶螨是一种世界性的重要害螨,地肤子提取物对其有很好的杀螨活性。本文通过2-DE技术分析了1mg·mL-1地肤子提取物处理后山楂叶螨蛋白质表达的变化。结果表明,处理组和对照的蛋白双向电泳银染图谱Pattern非常相似,对照胶和处理胶平均匹配率分别为43.8%和42.3%,对照胶有蛋白点1139±102个,处理胶有蛋白点1183±106个,这些蛋白多数集中于等电点5.0~9.0之间,分子量20~70ku的区域。通过PD-QUEST软件搜索和数据的统计分析,发现地肤子提取物触杀山楂叶螨会引起蛋白显著性变化(P〈0.05)。有262±23个蛋白点在表达量上有显著变化,其中142±13个点是2倍上调的点,另外120±11个点是下调的点,介于两者之间的点有237±21个。同时处理胶增加322±29个蛋白点,分子量主要集中在30~70ku,等电点集中在pH(5~6)和pH(7.5~9.0)之间。并推断试验中显著性变化的蛋白有可能是地肤子提取物的靶标蛋白。 The mite(Tetranychus viennensis Zacher) is a kind of important pests worldwide, and Kochia scoparia extracts have acaricidal activity on it. In this thesis, the protein of the mite which was treated by 1 mg .mL-1K, scoparia extract was analyzed by two-dimensional electrophoresis technology (2-DE). The results shows that the 2-DE silver staining pattern is very similar between the CK and the treatment, and the average matehingrates of the two groups are 43.8% and 42.3%, respectively. The CK gel has 1139 ±102 protein points while the treatment has 1183 ±106, and most of these proteins are concentrated in the isoelectric point of 5 - 9 and the molecular weight of 20 - 70 ku. The result also shows that the protein of the mite treated by K. scoparia extracts was significantly changed (P〈0.05) by PD-QUEST search and data analysis. There are 262 ± 23 protein spots with significant changes in expression, of which 142 ± 13 point is 2 fold up regulation of point, in addition to 120 ± 11 points down the point, between the two points is 237 ± 21. At the same time, the treatment increased by 322 ±29 protein spots, and the molecular weight of the proteins concentrated in 30-70ku and the isoelectric point on pH (5-6) and pH(7.5-9.0). So, we think that the protein which was significantly changed in the treatment gel may be the target protein of K. scoparia extracts.
