摘要
目的探讨胃转流术(gastric bypass,GBP)对2型糖尿病大鼠(T2DM)脂肪组织vaspin mRNA表达的影响。方法采用高脂饮食联合链脲佐菌素建立SD T2DM大鼠模型,随机分为T2DM-GBP组(A组)和T2DM-假手术组(B组),另取周龄、性别相匹配的正常SD大鼠,随机分为GBP组(C组)和假手术组(D组)。分别于术前及术后第4周和第8周检测大鼠的空腹血糖(FPG)水平,术后第8周处死大鼠,RT-PCR方法检测内脏脂肪组织vaspin mRNA表达。结果术后A组FPG水平呈进行性下降,术后第4周、第8周FPG水平均低于术前(P<0.05),B组、C组、D组内手术前及术后第4周、第8周FPG水平变化差异无统计学意义(P>0.05)。术后8周A组、C组、D组大鼠内脏脂肪组织vaspin mRNA表达水平比较,差异无统计学意义(P>0.05),而C组内脏脂肪组织vaspin mRNA表达水平分别高于A组、C组、D组,差异有统计学意义(P<0.05)。结论 GBP手术能显著降低T2DM大鼠血糖,可能与术后vaspin mRNA表达水平降低,胰岛素敏感性改善有关。
Objective To investigate the effects of gastric bypass( GBP) on expression of vaspin mRNA in omental adipose tissue of type 2 diabetes mellitus(T2DM) rats. Methods The models of T2DM rats were induced by Stre- ptozotocin and high-fat, randomly divided into 2 groups: T2DM-GBP group( A group), T2DM-sham operation( B group). 20 age-and sexmatched normal SD rats were randomly divided into 2 groups: GBP group(C group),and Sham operation(D group). Fasting plasma glucose (FPG)level was determined before operation and on week 4,8 after operation,respectively. At the same time,Rats omental adipose tissue was harvested eight weeks postoperatively and analyzed by using reverse transcription polymerase chain reaction (RT-PCR) to detect the vaspin mRNA expression. Results The FPG level progressively decreased in the A group,which was significant differences of the indicator between before operation and on week 4,8 after operation((P 〈 0. 05). Compared the indicator after operation with before operation,the FPG level was not statistically significant( P 〉 0. 05) in the B group,C group,or D group. Eight weeks after surgery, vaspin mRNA expression was no significant difference between A group, C group and D group. (P 〉 0. 05), which of the B group rats adipose tissue was higher than those in A group, C group,or D group. Conclusion GBP surgery can significantly decrease the fasting plasma glucose of T2DM rats, Maybe the low expression of vaspin mRNA in omental adipose tissue after GBP surgery play an important role in decreasing the FPG of T2DM rats.
