摘要 核磁共振技术(NMR)既可用于混合体系的定性分析,又可以用于其定量分析。在过去的几十年里,随着分析技术以及各种实验技术的迅速发展,基于核磁共振的定量分析方法已广泛应用于生物样本的分析。核磁共振定量分析技术应用于代谢组学,并成为定量代谢组学(quantitative metabolomics)研究中的重要手段。本文将论述这种新分析方法相比于传统方法的优势及不足之处,同时论述其研究过程中需考虑的重要因素以及其在代谢组学研究中的应用。 Nuclear magnetic resonance(NMR) spectroscopy can be used to both identify and quantify chemicals from complex mixtures. Over the last several decades, significant technical and experimental advances have made quantitative nuclear magnetic resonance(qNMR) a valuable analytical tool for quantitative measurements of a wide variety of samples. This particular approach is now being exploited to characterize the metabolomes of many different biological samples and is called quantitative metabolomics or targeted metabolic profiling. In this review, some of the strengths, limitations of NMR-based quantitative metabolomics will be discussed as well as the practical considerations necessary for acquisition with an emphasis on their use for bioanalysis. Recent examples of the application of this particular approach to metabolomics studies will be also presented.
