摘要
目的：建立以HPLC法测定两性霉素B滴眼液中两性霉素B含量的方法。方法：Shim—pack VP—ODS色谱柱（150mm×4．6mm，5μm），流动相为5mmol／LEDTA-乙腈-甲醇（30：25：60）；流速为1．2ml／min，检测波长为408nm，柱温为25℃。结果：两性霉素B在10～80μg／ml范围内线性关系良好（r=0．9999），平均回收率为98．51％。结论：本方法简单、准确，重现性、分离效果好，可用于两性霉素B滴眼液的质量控制。
Objective： To establish a method for the determination of amphotericin B in amphoteriein B eye drops by HPLC. Methods ： Chromatographic column ： shim - pack VP - ODS （ 150 mm× 4.6 mm,5 μm）, mobile phase ：5 mmol/L EDTA - acetoni- trile- methanol （30： 25： 60） , flow rate： 1.2 mlfmin, detection wavelength：408 nm, column temperature：25 ℃. Result：The calibration curve of assay is linear when the concentration of amphoteriein B is in the range of 10 - 80 μg/ml（ r = 0. 999 9 ） and the average recovery is 98.51%. Conclusion：The method is simple, curate with good reproducibility and resolution, and could be applied to content determination of amphoteriein B.
