摘要
目的:建立以HPLC法测定两性霉素B滴眼液中两性霉素B含量的方法。方法:Shim—pack VP—ODS色谱柱(150mm×4.6mm,5μm),流动相为5mmol/LEDTA-乙腈-甲醇(30:25:60);流速为1.2ml/min,检测波长为408nm,柱温为25℃。结果:两性霉素B在10~80μg/ml范围内线性关系良好(r=0.9999),平均回收率为98.51%。结论:本方法简单、准确,重现性、分离效果好,可用于两性霉素B滴眼液的质量控制。
Objective: To establish a method for the determination of amphotericin B in amphoteriein B eye drops by HPLC. Methods : Chromatographic column : shim - pack VP - ODS ( 150 mm× 4.6 mm,5 μm), mobile phase :5 mmol/L EDTA - acetoni- trile- methanol (30: 25: 60) , flow rate: 1.2 mlfmin, detection wavelength:408 nm, column temperature:25 ℃. Result:The calibration curve of assay is linear when the concentration of amphoteriein B is in the range of 10 - 80 μg/ml( r = 0. 999 9 ) and the average recovery is 98.51%. Conclusion:The method is simple, curate with good reproducibility and resolution, and could be applied to content determination of amphoteriein B.
