摘要
利用PCR-RFLP技术对549只西农萨能奶山羊和关中奶山羊的AGPAT6基因多态性与产乳性状的相关性进行了研究。结果表明,AGPAT6基因在P1、P2、P4与P10位点发现4个SNPs:5'UTR(g.152G>C)、内含子2(g.8124G>A)、外显子4(g.9263C>G)和3'UTR(g.16436G>A)。在P4位点,西农萨能羊GG基因型个体其产奶量显著高于CC型个体(P<0.05),在两个奶山羊品种的奶成分性状中,GG基因型个体的乳脂率显著高于CC型个体(P<0.05),GG型个体的乳蛋白含量极显著高于CC型个体(P<0.01),表明AGPAT6基因第4外显子不同基因型与山羊奶中的乳脂率和乳蛋白含量显著相关。因此,AGPAT6基因可以用于奶山羊产奶性状选育的重要候选基因。
The associations between AGPAT6 gene polymorphisms and milk yield trait were investiga- ted in 549 Xinong Saanen and Guanzhong dairy goat breeds using PCR--RFLP techniques. Four SNPs in- cluding 5"UTR(g. 152G〉C), intron2(g. 8124G〉A), exon4(g. 9263C〉G)and 3'UTR(g. 16436G〉A)were detected in four loci (P1, P2, P4 and P10) respectively. At P4 locus, individuals with GG genotype showed significant higher milk yield than those with CC genotype (P〈0.05) in Xinong Saanen breed. In- dividuals with GG genotype had significant higher milk fat (P〈0.05) and milk protein yield (P〈0.01) in two dairy goat breeds. The results indicate that there was significant association between different geno- types of exon 4 of AGPAT6 gene and milk fat and milk protein yield traits in dairy goats. In conclusion, the AGPAT6 gene can be considered as a candidate gene for milk traits breeding in dairy goats.
