摘要
目的探讨非酒精性脂肪肝中肝细胞固醇调节元件结合蛋白(SREBP-1C)与葡萄糖调节蛋白94(GRP-94)的表达变化,评价内质网应激蛋白对肝细胞脂质代谢的影响。方法应用半定量逆转录聚合酶链反应(RT-PCR)法和免疫组织化学法检测18例非酒精性脂肪肝患者和10例正常对照肝组织中SREBP-1C与GRP-94基因和蛋白表达,肝活检评分其炎症和纤维化。结果与正常对照组比较,非酒精性脂肪肝患者肝组织SREBP-1c与GRP-94基因和蛋白表达明显增强(P<0.01)。结论非酒精脂肪肝引起的肝脏损害可能与SREBP-1c和GRP-94表达有关,它们参与脂肪肝的发生,这可能是该病的发病机制之一。
【Objective】 To explore the expressions of sterol regulatory element binding protein-1C(SREBP-1C) and glucose-regulated protein 94(GRP-94) in human nonalcoholic fatty liver(NAFLD) and to evaluate the effects of endoplasmic reticulum stress proteins on hepatocytes lipids metabolism. 【Methods】 The expressions of SREBP-1C and GRP-94 in hepatic tissues were assessed in 24 patients with NAFLD and 8controls without liver disease using the RT-PCR and immunohistomistry. Liver biopsies were also graded for inflammation and fibrosis. 【Results】 A significant elevation in the expressions of SREBP-1C and GRP-94 in liver of NAFLD group were found compared to the control group(P〈0.01). 【Conclusion】 The excessive expressions of SREBP-1C and GRP-94 could participate in the development of nonalcoholic fatty liver. This information suggested that was involved in mechanism of nonalcoholic fatty liver.
