近红外光谱快速测定南板蓝根药材中水、醇浸出物的含量 被引量:1

Quantitative Analysis of Water Extract and Ethanol Extract in Rhizoma et Radix Baphicacanthis Cusiae by Near-infrared Spectroscopy

摘要 【目的】采用近红外光谱技术(NIRS)快速无损测定南板蓝根药材中水、醇溶性浸出物的含量。【方法】采用近红外漫反射光谱,结合偏最小二乘法(PLS)在OPUS/Quant 2中建立快速测定浸出物的定量模型,对模型进行验证,考察模型的可靠性。【结果】所建立的水、醇溶性浸出物校正模型决定系数(R2),交叉验证校正标准偏差(RMSECV)值分别为0.959、0.602和0.965、0.481;验证集的预测值与参考值的R2和RMSEP值分别为0.970、0.498和0.964、0.485;验证集的预测值与参考值的绝对误差均在±2%之间,预测值与参考值比较吻合,模型的建立较为成功。【结论】NIRS测定南板蓝根水、醇浸出物的含量方法准确、快速、简便,可作为南板蓝根质量检测的方法。 Objective To establish a rapid and non-destructive determination method for water extract and ethanol extract in Rhizoma et Radix Baphicacanthis Cusiae with near-infrared reflectance spectroscopy (NIRS) .Methods The quantitative model was built up by near-infrared diffuse reflectance spectrum combined with partial least square (PLS) in OPUS/Quant 2. We predicted the content of the unknown samples by the model and investigated the reliability of the model. Results The corrected model presented the coefficient of determination (R2) and root-mean-squares error of cross-validation (RMSECV) were 0.959, 0.602 for water extract, and 0.965, 0.481 for ethanol extract respectively. The RE and RMSEP of predicted value and reference value of validation set were 0.970, 0.498 for water extract and 0.964, 0.485 for ethanol extract respectively. The absolute error of predicted value and reference value of validation set was in the range of ± 2%. The predicted value was consistent with the reference value, which meant that the model had been established successfully. Conclusion The NIRS method is accurate, fast and practical, and it can be used as the quality testing method for Rhizoma et Radix Baphicacanthis Cusiae.
