摘要
应用膜片钳技术,以MN-9D神经细胞为材料,研究了溴氰菊酯及辛硫磷混剂的增效机理。试验表明,10^(-5)mol/L辛硫磷对Na^+通道电流抑制作用很小,并随作用时间延长而逐步恢复。加药1 min,Na^+电流抑制率为6.99%,10 min为3.65%。10^(-6)mol/L溴氰菊酯1 min抑制率为20.28%,10 min为21.43%。10^(-6)mol/L溴氰菊酯与10^(-5)mol/L辛硫磷混剂1 min抑制率为34.15%,10 min为36.69%,因此,混剂可增强对Na^+通道电流的抑制作用。通过Na^+电流数据、尾电流衰减时间常数统计分析表明,溴氰菊酯的修饰作用主要发生在关闭和静止状态的Na^+通道,减缓通道的打开,延长通道关闭或失活状态。
The synergitic mechanisms of deltamethrin and phoxim mixtures has been studied on mesencephalic originated dopaminergic cell line MN-9D by using the patch-clamp technique.It showed that 10^(-5)mol/L phoxim has little inhibition on sodium channel currents,and the inhibition will gradually disappear as time develops.For example,the inhibition rate of sodium channel current is 6.99% in one minute,3.65% in ten minutes after the 10^(-5)mol/L phoxim has been puffed.But deltamethrin has stronger inhibition on sodium channel current than the phoxim.The inhibition rate of 10^(-6) mol/L deltamethrin is 20.28% in one minute,21.43% in ten minutes.The inhibition rate of the mixture of 10^(-6) mol/L deltamethrin and 10^(-5) mol/L phoxim is 34.15% in one minute,36.69% in ten minutes.This reach the conclusion from the above experiment data that the effect of deltamethrin is to slow the opening,prolong the closed or inactivated state when the channel is in the closed or resting state.
