摘要
背景与目的:许多研究已经证实β-连环蛋白(beta-catenin)在多种人类肿瘤中有异常表达。为确定β-连环蛋白是否参与食管癌的发生发展,本实验检测β-连环蛋白在正常食管上皮及食管癌组织中的表达状态,研究其表达、定位与肿瘤分化程度及淋巴结转移的关系。方法:采用免疫组化SP法,检测52例食管癌及22例正常食管组织切片中β-连环蛋白的表达。结果:22例正常食管组织中,β-连环蛋白表达均显著位于细胞膜,而胞浆表达很少见;52例食管癌组织中,β-连环蛋白均呈现不同程度的胞浆表达,同时伴随细胞膜表达量减少甚至缺失,其中14例在细胞核内还可检出β-连环蛋白。另外,在有淋巴结转移的食管癌病例中,β-连环蛋白胞浆表达的阳性率明显高于无淋巴结转移的食管癌病例(P<0.05)。结论:β-连环蛋白在食管癌组织中表达异常较为普遍,主要表现在蛋白从细胞膜到细胞浆、到细胞核的转位,而且胞浆累积明显;这种改变与淋巴结转移率显著相关。
Background &Objective:Many reports have characterized the aberrant expression of beta-cateni n in diverse types of human cancer.To determine whether beta-catenin has possible roles in esophageal carcinoge nsis,we designed this study to detect the expression p attern ofβ-catenin in normal esophageal epith elium and esophageal cancer tissue,then to study the relevance of its expressio n and localization to tumor differetiation degree and lymph node metasta sis.Methods:By using immunohistochemical stainin g(SP method),the expression ofβ-catenin was detected in 22normal esophageal tissue slides and 52esophageal carcinomas.Results:In the 22normal esophageal tissue,β-catenin showed high intense expression at the membrane and low intense expre ssion at the cytoplasm.In contrast to the normal tissue,β-catenin was expressed in the cytoplasma in carcinoma with varied degrees,accompanied by less,or even lost expression at the membrane in cancer samples.In some cases,β-catenin could be detected in the nucleus.Moreover,the positive rate ofβ-catenin expression in cytoplasm was significantly higher in those patients with lymph node metastasis than patients without (P<0.05).Conclusions:The aberrant expression ofβ-catenin occurred frequently in the esophageal carcinoma,mainly inclu ding the translocation ofβ-catenin protein from membrane to th e cytoplasm and nucleus,and the accumulation ofβ-catenin in cytoplasma was overt.And the aberrant expression ofβ-catenin protein was statistically correlated to the lymph node metasta sis in esophageal cancer.
