缺氧对乳腺癌干细胞微球体培养及基因表达的影响 被引量:3

摘要 背景与目的三阴性乳腺癌侵袭转移的风险高、预后差。肿瘤转移的关键一是肿瘤干细胞,二是缺氧微环境,该研究拟探讨缺氧对乳腺癌干细胞MDA-MB-231微球体培养及基因表达的影响。方法分别在缺氧和常氧环境下进行乳腺癌干细胞的培养、富集、鉴定和分选,采用细胞因子抗体芯片检测缺氧与常氧乳腺癌干细胞基因表达的差异。结果缺氧条件下乳腺癌干细胞成球率及比例优于常氧条件,细胞因子抗体芯片显示,缺氧乳腺癌干细胞与常氧比有多个基因表达上调。结论缺氧微环境促进乳腺癌干细胞微球体的形成,并有多个基因表达上调。
