摘要
摇瓶中对转TNF -α基因鱼腥藻 712 0 (Anabaenasp .PCC712 0 ,pDC -TNF)混合培养条件进行了研究 ,在含蔗糖 9g/L ,NaNO3 2 .2 5g/L的BG - 11培养基混合培养时 ,得到最适培养条件接种量 5 %,光照强度为 10 0 0Lux ,光 /暗周期 (光照时间 /黑暗时间 ) 12h/ 12h ,温度 2 5~ 30℃ ,自然初始 pH值 ,10 0mL摇瓶装液量 4 0mL ,转基因鱼腥藻 15d生物量可达到 3g/L以上 ,可溶性蛋白含量接近 30 %,TNF表达水平大于 2 2 %,与自养相比 ,生物量增加 82 .0 6 %,表达水平提高38.79%。证明混合营养型培养是转rhTNF -α基因鱼腥藻 712 0实现高密度、高表达培养的途径。
The mixotrophic culture of recombinant Anabaena sp. PCC7120 harboring pDC-TNF was studied in shake flask and the optimum culture conditions were as follows. volume of inoculum was 5%, strength of illumination was 1000lux, temperature was 25~30℃, volume of media in 100ml flask was 40mL,and pH kept at 8.3 during the culture of recombinant Anabaena sp. PCC7120 in BG-11 media containing 9g/L sucrose and 2.25 g/L NaNO 3, the harvest biomass was over 3 g/L, the content of dissoluble protein and the expression of TNF was about 30% and more than 22%, respectively. The results showed that mixotrophic culture was the way for the recombinant Anabaena sp. PCC7120 to achieve high density and high expression.
