进出口食品中甲霜灵残留量免疫学快速检测方法的研究 被引量:6

摘要 将甲霜灵转化成甲霜灵酸 ,然后通过混合酸酐法连接到载体蛋白上 ,以此结合物免疫家兔 ,获得针对甲霜灵的特异性抗体 ,经间接ELISA测定其终点滴度 >1∶2 0 4 80 0 ,并建立了甲霜灵ELISA快速检测方法。抑制试验表明其检测范围在 1 953~ 2 50 0ng/mL之间 ,可用于植物产品中甲霜灵的定量检测。该方法与气相色谱检测符合率为 92 %。回收试验 :小麦平均回收率为 97 47%、土豆平均回收率为 99 45 %、大米平均回收率为97 0 %。交叉反应显示该抗体仅与甲霜灵、甲霜灵酸起反应 ,不与结构相似的毒草胺、甲草胺、异丙甲草胺、新燕灵和己酰甲草胺起交叉反应。本方法简便、快速、灵敏 。 Metalaxyl was acidified to its acid and the synthetized antigens were prepared by coupling the acid to carrying protein with the method of mixed anhydride and were used as immunogenes to immunize the rabbits.The titer of the antisera measured with indirect ELISA was>1∶ 204 800. The determination method for metalaxyl was set up.The range of detection was 1 953 ng/mL to 250 0 ng/mL,which was fit for the demand of quantitative determination of Metalaxyl in plant products.The coincidance rate between the test and the gas chromatography was 92%.The average recovery of fortified rice,wheat and tomato was 97%,97 47%and 99 45%respectively.Cross reactivity indicated that the antisera could only react with metalaxyl and its acid,but not with those matters whose structures are similar to metalaxyl including metolachlor,diethatyl ethyl,alachlor propachlor and benzoylprop ethyl.This method is simple,rapid and sensitive and fit for determination of metalaxyl residue in plant products.
