摘要
表达序列标记 (EST)是通过对cDNA文库克隆进行大规模测序所获得的 5′或 3′端序列 ,长度一般为 30 0~ 5 0 0bp ,可代表生物体某一时间内某组织中表达的基因 .本文就EST技术原理、概念最新发展及其在大规模快速克隆基因。
Expressed sequence tags(ESTs) are the 3′ or 5′ sequences obtained from large scale single-pass sequencing of cDNA, with the length of 300 to 500bp.ESTs can represent the transcriptional activity of a cell or tissue that include not only the identification of transcribed genes,but also abundance level.This technique is applied widely in cloning genes in large scale and in studying function of genes.It also has contributed a lots to the completion of the Human Genomic Plan (HGP) ahead of time.This article reviews the principle of this technique and the development of the concept-the module-EST.Its application in the electric cloning was also described.
