摘要
目的 建立成年小鼠骨髓间充质干细胞 (MSC)的分离和培养方法。方法 应用体外细胞培养技术将成年小鼠骨髓间充质干细胞自股骨、胫骨中分离、纯化和培养 ,用形态学鉴定培养的MSC ,用台盼蓝染色法及在光镜下观察分离培养的细胞和摄像纪录。结果 经培养存活的骨髓间充质干细胞呈长梭形 ,似成纤维样细胞 ,长宽比约为 2～ 3:1。
Objective To establish method of isolation and culture of mouse bone marrow derived mesenchymal stem cells(MSC).Methods MSC were isolated from the femurs and tibias bones and purified and in vitro culture, MSC quality was evaluated with morphology and trypan blue dye.States of MSC growing in culture were observed with electron microscope photography.Results Survival MSC were motionlessly attached to the bottom of culture plates,they were intact and fibroblast cell shaped, their length/width ratio was 2～3:1.Conclusions The method of isolation and culture of mouse bone marrow derived mesenchymal stem cells was successful.
