摘要
目的 研究 Caspase- 3在戊四氮 (pentylenetetrazole,PTZ)急性点燃大鼠海马组织中的表达情况。方法 通过腹腔注射戊四氮建立急性点燃大鼠模型 ,运用逆转录聚合酶链式反应技术分析 Caspase- 3m RNA在癫痫发作后的表达情况。结果 癫痫发作后海马组织 Caspase- 3m RNA的表达水平早期即出现升高 (8h组与对照组相比 ,P<0 .0 5 ) ,且持续较长一段时间 (5 d组与对照组相比 ,P<0 .0 5 )。结论 Caspase- 3可能参与了癫痫后海马神经元的凋亡过程。
Objective To investigate the expression of Caspase 3 in the hippocampus of acutely pentylenetetrazole-kindled rats. Methods Acutly kindled rat model was made by intraperitoneal injection of pentylenetetrazole. The hippocampal expression of Caspase 3 mRNA was showed through reverse transcription polymerase chain reaction (RT PCR) method. Results Caspase 3 mRNA was significantly up regulated in hippocampal neurons following PTZ induced epilepsy and existed for several days(at least from 8h to 5d after epilepsy). Conclusion The result indicates that Caspase 3 may be activated at transcriptional level during neuronal apoptosis after epilepsy.
